Abstract The origins of resting-state functional MRI (rsfMRI) signal fluctuations remain debated. Recent evidence shows coupling between global cortical rsfMRI signals and cerebrospinal fluid inflow in the fourth ventricle, increasing during sleep decreasing with Alzheimer’s disease (AD) progression, potentially reflecting brain clearance mechanisms. However, existence more complex brain-ventricle modes their relationship to cognitive decline remains unexplored. Analyzing 599 minimally-preprocessed scans from 163 elderly participants across AD spectrum, we identified distinct that differentiate groups correlate scores. Beyond known anti-phase ventricles —more frequent cognitively normal controls— discovered additional where specific areas temporarily align ventricle signals. At level, these form canonical networks, such as Default Mode Network, which occurs less or Frontoparietal correlates positively memory direct link challenges common practice removing CSF components analyses questions origin forming may reflect region-specific patterns. These findings provide new insights into mechanisms network dysfunction neurodegenerative diseases.

Analysis and interpretation of studies on cognitive affective dysregulation often draw upon the network paradigm, especially Triple Network Model, which consists default mode (DMN), frontoparietal (FPN), salience (SN). DMN activity is primarily dominant during leisure self-monitoring processes. The FPN peaks task involvement exertion. Meanwhile, SN serves as a dynamic “switch” between FPN, in line with demand. In domains, dysfunctions involving are connected to broad spectrum deficits maladaptive behavioral patterns variety clinical disorders, such depression, insomnia, narcissism, PTSD (in case hyperactivity), chronic pain, anxiety, high degrees neuroticism, schizophrenia, epilepsy, autism, neurodegenerative illnesses, bipolar disorder hypoactivity). We discuss neurological data from various research domains present an integrated perspective indicating that these conditions can be associated widespread disruption predictive coding at multiple hierarchical levels. delineate fundamental ideas brain paradigm contrast them conventional modular method first section this article. Following this, we outline interaction model key functional networks highlight recent coupling SN-related impairments.

Controllability analysis is an approach developed for evaluating the ability of a brain region to modulate function in other regions, which has been found be altered major depressive disorder (MDD). Both symptoms and cognitive impairments are prominent features MDD, but case-control differences controllability between MDD controls can not fully interpret contribution both clinical cognition linked patterns among them MDD.

The brain is always intrinsically active, utilizing energy at high rates, while moving between global functional modes. Awake modes are tied to corresponding behavioral states. During goal-directed behavior, the enters an action-mode of function. In action-mode, arousal heightened, attention focused externally, and action plans created, converted movements, continuously updated based on relevant feedback, such as pain. Here, we synthesize classical recent human animal evidence that brain’s created maintained by network (AMN), which had previously identified named cingulo-opercular (CON) its anatomy. Controlling accounts for large variety functions associated with AMN, increasing arousal, processing instructional cues, task general initiation transients, sustained goal maintenance, planning, sympathetic drive (e.g., connectivity adrenal medulla) controlling physiology internal organs, well action-relevant bottom-up signals pain, errors viscerosensation. mode space, AMN-generated antipole default-mode self-referential, emotional, memory processing.

Decades of neuroscience research has shown that macroscale brain dynamics can be reliably decomposed into a subset large-scale functional networks, but the specific spatial topographies these networks and names used to describe them vary across studies. Functional connectivity (FC) is conventionally defined by measuring the similarity between brain signals from two regions. The technique has become widely adopted in analysis of functional magnetic resonance imaging (fMRI) data, where it provided cognitive neuroscientists with abundant information on how regions interact to support complex cognition. However, past decade notion "connectivity" expanded both complexity and heterogeneity its application neuroscience, resulting greater difficulty interpretation, replication, cross-study comparisons. In this paper, we begin canonical notions then introduce recent methodological developments that either estimate some alternative form or extend analytical framework, hope bringing better clarity for neuroscience researchers.

Abstract Approaching the 30th anniversary of discovery resting state functional magnetic resonance imaging (rsfMRI) connectivity, we reflect on impact this neuroimaging breakthrough field child and adolescent psychiatry. The study intrinsic brain architecture that rsfMRI affords across a wide range ages abilities has yielded numerous key insights. For example, now know many neurodevelopmental conditions are associated with more widespread circuit alterations multiple large-scale networks than previously suspected. emergence population neuroscience effective data-sharing initiatives have made large datasets publicly available, providing sufficient power to begin identify brain-based subtypes within heterogeneous clinical conditions. Nevertheless, several methodological theoretical challenges must still be addressed fulfill promises personalized In particular, incomplete understanding physiological mechanisms driving developmental changes in connectivity remains an obstacle further progress. Future directions include cross-species multimodal investigations illuminate such mechanisms. Data collection harmonization efforts span countries diverse cohorts urgently needed. Finally, incorporating naturalistic fMRI paradigms as movie watching should priority for future research efforts.
