The varied nature makes it difficult point specific cause the syndrome. However, current literature agrees that inflammation cornea conjunctiva plays major role in its pathogenesis. Therapies targeted toward shown success treatment DED. purpose this review provide an overview prevalence inflammatory pathophysiology discussion available anti-inflammatory therapies including following: Nonsteroidal drugs, corticosteroids, other hormonal therapies, nonsteroidal immunomodulators, biological tear replacement, antibiotics, dietary supplements, tea tree oil, intense pulsed light.

Homeostasis of the lacrimal functional unit is needed to ensure a well-regulated ocular immune response comprising innate and adaptive phases. When system excessively stimulated and/or immunoregulatory mechanisms are disrupted, balance between phases dysregulated chronic surface inflammation can result, leading dry eye disease (DED). According Tear Film Ocular Surface Society Dry Eye Workshop II definition, DED multifactorial disorder characterized by impairment loss tear homeostasis (hyperosmolarity), discomfort or pain, neurosensory abnormalities. Dysregulated responses result in damage, which further contributing factor pathology. Several therapeutics available break vicious circle prevent progression, including immunosuppressive agents (steroids) immunomodulators (cyclosporine lifitegrast). Given inflammatory nature DED, each these commonly used clinical practice. In this study, we review immunopathology molecular cellular actions current topical inform decision making.

Prolonged and continuous daily use of digital screens, or visual display terminals (VDTs), has become the norm in occupational, educational, recreational settings. An increased global dependence on VDTs led to a rise associated complaints, including eye strain, ocular dryness, burning, blurred vision, irritation, name few. The principal causes for VDT-associated discomfort are abnormalities with oculomotor/vergence systems dry (DE). This review focuses latter, as advances research have identified symptomology surface parameters that shared between prolonged VDT users DE, particularly evaporative subtype. Several mechanisms been implicated blink anomalies, damaging light emission from modern devices, inflammatory changes. presence preexisting DE also explored an inciting exacerbating factor. We associations screens damage, therapeutic options, hoping raise awareness this entity goal reducing morbidity economic impact screen-associated disability.

Exosomes, nanosized extracellular vesicles of 30–150 nm, are shed by almost all cell types. Bearing proteins, lipids, RNAs, and DNAs, exosomes have emerged as vital biological mediators in cell-to-cell communication, affecting a plethora physiological pathological processes. Particularly, mounting evidence indicates that immunologically active can regulate both innate adaptive immune responses. Herein, we review recent advances the research several immune-mediated eye diseases, including Sjögren's syndrome (SS) dry eye, corneal allograft rejection, autoimmune uveitis, age-related macular degeneration (AMD). Additionally, discuss potential novel biomarkers drug delivery for diagnosis treatment diseases.

Abstract Ocular injuries and their complications represent the most common causes of visual impairment. For ocular surgery, there is an unmet need for highly transparent bioadhesives with superior adhesion, biocompatibility, regenerative properties. Herein, a novel high‐transparent bioadhesive hydrogel composed gelatin methacryloyl (GelMA) dopamine methacrylamide (DMA) developed by in situ oxidative free‐radical polymerization. This overcomes fundamental weakness mussel‐inspired adhesive copolymers clinical practice combining multiple favorable properties, including high light transmission, mechanical strength, biocompatibility. DMA significantly enhances corneal epithelial cells proliferation, migration on GelMA, prevents accumulation reactive oxygen species (ROS) cells. In rabbit models conjunctiva transplantation, able to decrease inflammatory response fibrosis formation induced suture surgical trauma. addition, stromal defect model reveals that Gel/DMA could effectively seal defects, accelerates re‐epithelialization, promotes wound healing. Thus, given advantages bioactivity simple preparation, represents promising strategy suture‐free repair.

Abstract The present study was set out to address the therapeutic efficacy of human adipose tissue stem cells derived extracellular vesicles (hADSC-Evs) in a mouse model dry eye disease and investigate underlying mechanisms involved. hADSC-Evs drops were topically administered mice that subjected desiccating stress (DS). Clinical parameters ocular surface damage assessed with fluorescein staining, tear production PAS staining. For vitro studies, cell viability assay TUNEL staining performed corneal epithelial (HCECs) treated under hyperosmotic media. In addition, immunofluorescent Real-time PCR (qRT-PCR) Western blots used evaluated NLRP3, ASC, caspase-1, IL-1β expression levels. Compared vehicle control mice, topical showed decreased defects, increased production, goblet loss, as well reduced inflammatory cytokines production. vitro, could protect HCECs against stress-induced apoptosis. Mechanistically, treatment suppressed DS induced rises NLRP3 inflammasome formation, caspase-1 activation maturation. conclusion, effectively suppress response alleviate disease.
