Senescent chondrocytes, which are increased in osteoarthritic (OA) cartilage, promote cartilage defects and the senescent knee microenvironment by inducing senescence to surrounding normal chondrocytes secreting senescence-associated secretory proteins. Many studies have used mesenchymal stem cells (MSCs) treat OA, but MSC treatment remains challenging for clinical application owing quality control, engraftment, fibrocartilage regeneration. Here, rather than relying on direct regeneration of MSCs, we present a novel strategy suppress OA MSC-mediated chondrocyte targeting via paracrine activity thereby improving microenvironment. First, enable control umbilical cord priming MSCs supplementing human platelet lysate (hPL) greatly enhanced functions increasing cellular glutathione levels throughout serial passaging. Intra-articular injection primed successfully suppressed progression accumulation without Indirect coculture with using transwell ameliorated phenotypes suggesting rejuvenation. Based secretome analysis, identified insulin-like growth factor 2 (IGF2) as key component that induces rejuvenation MSCs. The effects IGF2 act through autophagy activation up-regulation autophagy-related gene expression autophagic flux. To cross-validate secreted vivo, knockdown substantially abolished its therapeutic efficacy rabbit model. Collectively, these findings demonstrate hPL supplementation enables levels. mechanism was IGF2, activating autophagy.

Aging is a process characterized by tissue degeneration, increased susceptibility to chronic degenerative diseases, infections, and the appearance of neoplasms, which leads disability reduction in length quality life. This phenomenon result convergence multiple processes, including mitochondrial dysfunction, fibrosis, inflammation, dysregulation cell death immunosenescence. These processes have as their point an increase production ROS. Avocado oil (Persea americana Mill.) contains diverse array bioactive compounds, oleic acid, phytosterols, chlorophylls, xanthones, xanthines, carotenoids. compounds capacity modulate excessive ROS, thereby reducing progression age-related diseases extending lifespan experimental models aging. In addition, several studies demonstrated efficacy avocado mitigating hypertension; insulin resistance; diabetes; non-alcoholic liver disease; such hearing loss, cognitive decline, neurodegeneration, impaired wound healing. light these findings, it hypothesized that promising agent capable promoting healthspan later stages life owing its direct antioxidant actions activation pathways enhance endogenous levels.

Background and purpose: Ponatinib, a tyrosine kinase inhibitor (TKI) leads to hypertension; however, the mechanisms remain elusive. We aimed investigate whether lysine acetyltransferase 7 (KAT7), key regulator of cellular senescence that is closely associated with cardiovascular diseases, involves in ponatinib-induced hypertension. Methods results: After administering ponatinib Sprague–Dawley (SD) rats for 8 days, we measured blood pressure, vasodilation, endothelial function using tail-cuff plethysmography, isometric myography, Total NO Assay kit, respectively. The results indicated increased impaired endothelium-dependent relaxation (EDR), caused injury cells SD rats. Furthermore, PCR Western blot experiments demonstrated an upregulation KAT7 expression rat mesenteric artery (MAECs) following treatment. To further study role hypertension, divided into four groups: control, ponatinib, WM-3835 (a inhibitor), plus WM-3835. Notably, administration significantly improved hypertension EDR dysfunction Mechanistically, over-expression (OE-KAT7) MAECs led inflammation, phenomena were also observed arteries ponatinib-treated exposed ponatinib. However, mitigated these detrimental effects both vivo vitro experiments. Additionally, OE-KAT7 treatment induced H3K14 acetylation (H3K14ac), elevating recruitment H3K14ac p21 promoter. Moreover, BAY 11-7085, nuclear factor (NF)-κB inhibitor, potently alleviated accumulation IL-6 IL-8, as well cell by overexpression. Conclusion: Our data indicate elevation inflammatory responses through NF-κB signaling pathway, subsequently vasotoxicity

The retina is part of the central nervous system specialized for vision. Inherited retinal diseases (IRD) are a group clinically and genetically heterogenous disorders that lead to progressive vision impairment or blindness. Although each disorder rare, IRD accumulatively cause blindness in up 5.5 million individuals worldwide. Currently, pathophysiological mechanisms not fully understood there limited treatment options available. Most caused by degeneration light-sensitive photoreceptors. Genetic mutations abrogate structure and/or function photoreceptors visual followed loss In healthy retina, structurally functionally interact with pigment epithelium (RPE) Müller glia (MG) maintain homeostasis. Multiple photoreceptor as major phenotype RPE- MG-associated genes. Recent studies also reveal compromised MG RPE ubiquitously expressed ciliary Therefore, could be direct consequence gene secondary dysfunction their interaction partners retina. This review summarizes photoreceptor-RPE/MG supporting functions discusses how disruption these processes degeneration, an aim provide unique perspective pathogenesis paradigm. We will first describe biology then discuss between MG/RPE well implications disease pathogenesis. Finally, we summarize recent advances therapeutics targeting RPE.
