Abstract In the background of limited studies on noncoding RNAs in Drosophila oogenesis, we show developmentally active hsr ω lncRNA gene to be essential oogenesis and ovulation. The near-null 66 females, ovaries with down- or up-regulated display varyingly perturbed including fewer ovarioles, high apoptosis, poor actin nuclear-cage (stage 10), low Cut levels late chambers and, finally ovulation block. Restoration normal following targeted expression ω-RH transcript confirmed mis-function underlie these defects. Genetic interaction showed varying modulation ovarian defects mis-expression Cut, TBPH/TDP-43 Caz/dFUS hnRNPs by altered levels. Dietary supplement ecdysone which have reduced titer, substantially restored oogenesis. Our results for first time that multiple producing gene, interacts diverse other regulatory molecules, As expected a key roles dynamics various hnRNPs, interactions between down upregulated transcripts regulators are not linear. Summary Statement critically impacts at steps through intra- inter-organ signaling.

In Drosophila melanogaster and other insects, the seminal fluid proteins (SFPs) male sex pheromones that enter female with sperm during mating are essential for fertility induce profound post-mating effects on physiology. The SFPs in D. taxa include several members of large gene family known as odorant binding (Obps). Work has shown some Obp genes highly expressed antennae can mediate behavioral responses to odorants, potentially by carrying these molecules receptors. These observations have led hypothesis Obps might act molecular carriers or compounds important fertility, though functional evidence any species is lacking. Here, we used genetics test role seven response (PMR). We found Obp56g required induction PMR, whereas six dispensable. males lacking fail form a plug mated female's reproductive tract, leading ejaculate loss reduced storage, likely due its expression ejaculatory bulb. also examined evolutionary history genes, studies documented rapid evolution turnover SFP across taxa. extensive lability copy number positive selection acting two Obp22a Obp51a. Abstract Studies of allele-specific expression in interspecific hybrids have provided important insights into gene-regulatory divergence and hybrid incompatibilities. Many such investigations Drosophila used transcriptome data from complex mixtures many tissues or gonads, however, regulatory may vary widely among species, sexes, tissues. Thus, we lack sufficiently broad sampling to be confident about the general biological principles divergence. Here, seek fill some these gaps literature by characterizing evolution misexpression a somatic male sex organ, accessory gland, F1 between melanogaster D. simulans. The gland produces seminal fluid proteins, which play an role female fertility subject adaptive due male–male male–female interactions. We find that trans differences are relatively more abundant than cis, contrast most literature, though large effect-size rare. Seminal protein genes significantly elevated levels tend regulated through both cis limited (over- underexpression relative parents) this organ compared other studies. As previous studies, male-biased overrepresented misexpressed much likely underexpressed. ATAC-Seq show chromatin accessibility is correlated with species expression. In species with internal fertilization, sperm, and seminal fluid are transferred from male to female during mating. While both sperm contain various types of molecules, including RNA, the role most these molecules in coordination fertilization or other possible functions is poorly understood. Drosophila, exosomes accessory gland, which produces fluid, females, but their potential cargoes have not been described. Moreover, while RNA composition has described several mammalian species, little work on this problem occurred Drosophila. Here we use single nucleotide polymorphism differences between males females a set highly inbred lines D. melanogaster, transcriptome data reproductive tract, testis, investigate origin, vs female, isolated 3 tract organs, receptacle spermatheca, store parovaria, does not. We find that mated carry male-derived transcripts many genes, those markers gland known proteins. Our observations also support idea intact can be storage organs.

Seminal fluid proteins (SFPs) are key factors in sexual reproduction and transferred to females during mating with sperm. SFPs have a nutritional value because they protect activate sperm storage release optimize fecundity. Multiple matings promote ovipositioning several insect species. Therefore, insects may obtain more SFP through multiple maximize reproduction, but this process has not yet been clearly confirmed. Here, the relationship between Ophraella communa (Coleoptera: Chrysomelidae), biological control agent of common ragweed Ambrosia artemisiifolia (Asterales: Asteraceae), was studied. significantly increased female fecundity ovary egg deposition. Carboxypeptidase B (OcCpb) carbonic anhydrase (OcCa) genes were identified as putative O. showed strong male-biased expression. Additionally, OcCpb OcCa expression upregulated bursa copulatrix compared that virgin females, their gradually declined after copulation. Furthermore, knockdown males led decrease control. The reproductive tract mated dsRNA-treated dissected observed and, notably, ovaries produced fewer eggs. These data suggest play regulatory roles communa.

Long matings are abundant in insects despite the range of costs involved. The causes and consequences evolution long remain an interesting problem for behavioural ecologists. We studied Parthenium beetle (Zygogramma bicolorata), which is known to show extraordinarily copulations. Our detailed investigation showed, first time, that mounting this species entails repeated copulation. Each these copulations preceded succeeded by a leg-rubbing behaviour. found same characteristic pattern laboratory semi-natural conditions. conducted series interrupted assays examine fitness duration mounting. tested multiple hypotheses regarding transfer sperm non-sperm components, function leg rubbing modulating female copulatory Sperm fertility did not exhibit linear increase with number There was also no evidence nutrient males. Genes involved in reproduction often evolve rapidly due to postcopulatory sexual selection (PCSS) driven by male-male competition and male-female conflict, but the impact of PCSS on gene expression has been under-explored. Further, though multiple tissues contribute male reproductive success, most studies have focused testes. To explore influence mating system variation Peromyscus tract, we generated RNAseq libraries quantified testis, seminal vesicle, epididymis, liver wild-caught monogamous P. californicus polygynandrous boylii maniculatus males tested for positive relaxed Peromyscus-expressed genes using published rodent genomes. We identified 3,627 mating-system associated differentially expressed (MS-DEGs), where shifted same direction relative californicus. Gene was strongly with behavior vesicles, 89% were MS-DEGs, including two key fluid proteins, Svs2 Pate4. Though showing evidence or differential did not overlap, 203 MS-DEGs showed selection. Additionally, fourteen under tree-wide convergent Microtus ochrogaster, a distantly related species.
