With the recent description of molecular and cellular characteristics that enable acquisition both core new hallmarks cancer, consequences transcription factor dysregulation in scheme has become increasingly evident. Dysregulation or mutation factors long been recognized development cancer where alterations these key regulatory molecules can result aberrant gene expression consequential blockade normal differentiation. Here, we provide an up-to-date review involvement dysregulated networks with most recently reported enabling characteristic properties. We present some illustrative examples impact factors, specifically focusing on phenotypic plasticity, non-mutational epigenetic reprogramming, polymorphic microbiomes, senescence. also discuss how insights into is contributing to addressing current therapeutic challenges.

Abstract Background Microbial dysbiosis has emerged as an important element in the development and progression of various cancers, including breast cancer. However, microbial composition from healthy individuals, even relative to risk developing cancer, remains unclear. Here, we performed a comprehensive analysis microbiota normal tissue, which was analyzed relation tumor adjacent tissue. Methods The study cohorts included 403 cancer-free women (who donated tissue cores) 76 cancer patients and/or samples). Microbiome profiling obtained by sequencing nine hypervariable regions 16S rRNA gene (V1V2, V2V3, V3V4, V4V5, V5V7, V7V9). Transcriptome also on 190 samples. Breast score assessed using Tyrer-Cuzick model. Results V1V2 amplicon resulted more suitable for microbiome identified Lactobacillaceae ( Firmicutes phylum), Acetobacterraceae , Xanthomonadaceae (both Proteobacteria phylum) most abundant families breast. Ralstonia both tumors histologically tissues malignant tumors. We conducted correlation between known factors. Abundances bacterial taxa Acetotobacter aceti Lactobacillus vini, paracase i, Xanthonomas sp. were associated with age p < 0.0001), racial background parity 0.0001). Finally, transcriptome showed enrichment metabolism- immune-related genes vini paracasei whereas presence linked dysregulation involved carbohydrate metabolic pathway. Conclusions This defines features thus providing basis understand cancer-related dysbiosis. Moreover, findings reveal that lifestyle factors can significantly affect composition.

Breast cancer (BC) is the most common in women United States. There has been an increasing incidence and decreasing mortality rate of BC cases over past several decades. Many risk factors are associated with BC, such as diet, aging, personal family history, obesity, some environmental factors. Abstract Breast cancer was the most commonly diagnosed worldwide in 2020. Greater understanding of factors which promote tumour progression, metastatic development and therapeutic resistance is needed. In recent years, a distinct microbiome has been detected breast, site previously thought to be sterile. Here, we review clinical molecular relevance oral anaerobic bacterium Fusobacterium nucleatum breast cancer. Observational epidemiological studies suggested an association between the gut microbiota and breast cancer, but it remains unclear whether causally influences risk of cancer. We employed two-sample Mendelian randomization (MR) analysis to investigate this association. We used summary statistics microbiome from a genome-wide study (GWAS) 18,340 individuals in MiBioGen study. GWAS for overall cancer hormone receptor subtype-specific analyses were obtained UK Biobank FinnGen databases, totaling 400,000 individuals. The inverse variance-weighted (IVW) MR method was examine causal relationship its subtypes. Sensitivity conducted using maximum likelihood, MR-Egger, pleiotropic residual sums outliers methods. The IVW estimates indicated that increased abundance Genus_Sellimonas is associated with ER+ [odds ratio (OR) = 1.09, p 1.72E-04, false discovery rate (FDR) 0.02], whereas Genus_Adlercreutzia protective against (OR 0.88, 6.62E-04, FDR 0.04). For Her2+ Genus_Ruminococcus2 decreased 0.77, 4.91E-04, 0.04), Genus_Erysipelatoclostridium 1.25, 6.58E-04, No evidence heterogeneity or horizontal pleiotropy found. Our revealed microbiota-mammary axis, providing important data supporting potential use as candidate target prevention, diagnosis, treatment.

Globally, breast cancer is the leading cause of incidence and mortality among all female cancers. Hereditary factors only account for 5-10% cancers, highlighting importance non-hereditary factors, such as obesity. The increasing prevalence obesity underscores need to understand its contribution risk. Multiple mechanisms may mediate pro-carcinogenic effects obesity, including altered adipokine levels, local systemic inflammation, disruption insulin insulin-like growth factor signaling, increased estrogen alterations microbiome. In this review, we focus on link between gut microbiome risk in context First, discuss how influences Next, describe effect carcinogenesis, underlying molecular mechanisms. Finally, review preclinical data interactions host bacteria, current challenges study obesity-microbiome connection, future perspectives field.

Abstract Estrogen receptor-negative [ER(−)] mammary cancer is the most aggressive type of breast (BC) with higher rate metastasis and recurrence. In recent years, dietary prevention BC epigenetically active phytochemicals has received increased attention due to its feasibility, effectiveness, ease implementation. this regard, combinatorial phytochemical intervention enables more efficacious inhibition by simultaneously targeting multiple tumorigenic pathways. We, therefore, focused on investigation effect sulforaphane (SFN)-rich broccoli sprouts (BSp) withaferin A (WA)-rich Ashwagandha (Ash) combination in estrogen using transgenic mice. Our results indicated that BSp + Ash treatment significantly reduced tumor incidence growth (~ 75%) as well delayed 21%) latency when compared control was statistically effective suppressing single or intervention. At molecular level, upregulated suppressors (p53, p57) along apoptosis associated proteins (BAX, PUMA) BAX:BCL-2 ratio. Furthermore, our result an expressional decline epigenetic machinery HDAC1 DNMT3A tissue because treatment. Interestingly, we have reported synergistic interactions between impacted both phenotype expression RNA-seq analysis also demonstrated a transcriptome-wide reshuffling genes cell-signaling pathways, transcription factor activity regulations combined administration. addition, discovered alteration gut microbial composition change Overall, supplementation can prevent ER(−) through enhanced suppression, induction gene possibly influencing cell signaling regulation reshaping microbiota.

Abstract Breast cancer (BC) is one of the most diagnosed cancers in women. Based on histological characteristics, they are classified as non-invasive, or situ (tumors located within milk ducts lobules) and invasive. BC may develop from carcinomas over time. Determining prognosis predicting response to treatment essential tools manage this disease reduce its incidence mortality, well promote personalized therapy for patients. However, half cases not associated with known risk factors. In addition, some patients resistance relapse.
