ABSTRACT Emerging preclinical data suggests dietary interventions, including intermittent fasting, may play a key role in altering cancer progression. In feasibility trial monitoring cellular, clinical, and qualitative changes, ten patients with chronic lymphocytic leukemia followed time-restricted eating for three months, five six months. Seven of fifteen participants (47%) experienced decrease or stabilization malignant lymphocyte counts on eating, while accumulation slowed (33%) had no effect (20%). A reduction were accompanied by an unexpected decline cellular autophagy lymphocytes. Metabolite profiling identified microbial-derived bile acid metabolites, glycoursodeoxycholic, taurochenodeoxycholic, glycolithocholic ursodeoxycholic acid, short-chain fatty acids, dehydrolithocholic apocholic that changed eating. Participant quality life improved during These results connect shifts autophagy, tumor progression, life.

Abstract Objective To investigate the association of timing, frequency, and food quality night eating with all-cause, cancer, diabetes mortality. Methods This study included 41,744 participants from US National Health Nutrition Examination Survey (2002–2018). Night information was collected by 24-h dietary recall exposures were eating. Food assessed latent class analysis. The outcomes mortality, which identified Death Index International Classification Diseases 10th Revision. Adjusted hazard ratios [aHR] 95% confidence intervals [CI] computed Cox regression. Results During a median follow-up 8.7 years, 6066 deaths documented, including 1381 cancer 206 diabetes. Compared no (eating before 22:00), later timing associated higher risk all-cause mortality (each P -trend <0.05) rather than highest being 00:00–1:00 (aHR 1.38, CI 1.02–1.88) 23:00–00:00 2.31, 1.21–4.40), respectively. However, increased risks not observed for 22:00-23:00. Likewise, one time or over frequency < 0.05). That further in high-dietary-energy-density group (all-cause mortality: aHR 1.21 [95% 1.06–1.38]; 1.97 1.13–3.45]), but low-dietary-energy-density group. Finally, correlation analysis found positive associations glycohemoglobin, fasting glucose, OGTT. Conclusions mortality; however, reduction excess when 23:00 foods.

The circadian system is composed by a central hypothalamic clock at the suprachiasmatic nuclei (SCN) that communicates with peripheral oscillators for daily coordination of behavior and physiology. SCN entrain to environmental 24-h light-dark (LD) cycle drive rhythms internal synchronizers such as core body temperature, hypothalamic-hypophysary hormones, sympathetic/parasympathetic activity, well behavioral feeding-fasting rhythms, which supply signals setting molecular clocks tissues. Steady phase relationships between keep homeostatic processes microbiota/microbiome composition/activity, metabolic supply/demand, energy balance, immunoinflammatory process, sleep amount quality, psychophysiological stress, etc. Indeed, risk health alterations increase when these are chronically changed prompting disruption (CD), occurring after sudden LD changes (so-called jet-lag), or due activity/feeding-rest/fasting rhythm respect cycles (as humans subjected nightwork, restricting food access rest in mice). Typical pathologies observed animal models CD epidemiological studies include syndrome, type-2 diabetes, obesity, chronic inflammation, cancer, disruption, decrease physical cognitive performance, mood, among others. present review discusses different aspects physiological dysregulations having altered potential translation human health.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has surpassed the hepatitis B virus and C as leading cause of chronic in most parts Western world. MASLD (formerly known NAFLD) encompasses both simple steatosis more aggressive metabolic steatohepatitis (MASH), which is accompanied by inflammation, fibrosis, cirrhosis, ultimately can lead to hepatocellular carcinoma (HCC). There are currently very few approved therapies for MASH. Weight loss strategies such caloric restriction ameliorate harmful effect MASH inhibit HCC; however, it difficult implement maintain daily life, especially individuals diagnosed with HCC. In this study, we tested a time-restricted feeding (TRF) nutritional intervention mouse models We show that TRF abrogated dysregulation induced diet without any calorie or weight loss. improved insulin sensitivity reduced hyperinsulinemia, steatosis, fibrosis. Importantly, inhibited tumors two obesity-driven Our data suggest likely be effective abrogating HCC warrant further studies eating humans who at higher risk developing

Circadian oscillations of several physiological and behavioral processes are an established process in all the organisms anticipating geophysical changes recurring during day. The time-keeping mechanism is controlled by a transcription translation feedback loop involving set well-characterized factors. synchronization cells, at organismal level brain central clock, can be mimicked vitro, pointing to notion that cells endowed with autonomous system. Metabolism undergoes circadian control, including mitochondrial terminal catabolic pathways, culminating under aerobic conditions electron transfer oxygen through respiratory chain coupled ATP synthesis according oxidative phosphorylation chemiosmotic mechanism. In this study, we expanded upon previous isolated observations utilizing multiple cell types, employing various protocols different methodologies measure consumption rates simulating metabolic stressors. results obtained clearly demonstrate activity rhythmic tested regardless their individual proficiency, indicating phenomenon generalized. However, notably, while primary types exhibited similar profiles, cancer-derived lines displayed highly heterogeneous changes. This observation confirms on one hand dysregulation control metabolism observed cancer, likely contributing its development, other underscores necessity personalized chronotherapy, which necessitates detailed characterization cancer chronotype.

Abstract Background Basal metabolic rate (BMR) reflecting the body's energy expenditure at rest, may be a crucial indicator of accelerated aging. However, there is lack evidence for causal effect BMR on We aimed to apply Mendelian randomization (MR) and mediation analysis explore relationships between epigenetic age acceleration. Method Genetic variants linked (GrimAge, PhenoAge, HannumAge HorvathAge) acceleration were extracted from genome-wide association study datasets. MR primarily utilizes Inverse variance weighted (IVW), followed by range sensitivity analyses validate robustness our findings. Results The univariable demonstrated significant GrimAge (IVW: Beta 0.38 [95% CI 0.12 0.64]), PhenoAge accelerations 0.91 [95%CI 0.67 1.45]), but not in reverse direction. In multivariate adjusting body mass index (BMI), hypertension, type 2 diabetes mellitus (T2D), physical activity, smoking, alcohol intake frequency, total bioavailable testosterone levels, we found that effects both dependent BMI, also T2D, levels Mediation indicated 7.24%, 16.29%, 6.95%, 5.36% was mediated activity intake. Conclusion A positive unidirectional acceleration, which BMI.
