Abstract The pro‐inflammatory cytokine interleukin 17 (IL‐17), that is mainly produced by Th17 cells, has been recognized as a key regulator in multiple sclerosis (MS) and experimental autoimmune encephalomyelitis (EAE). Reactive astrocytes stimulated proinflammatory cytokines including IL‐17 are involved blood brain barrier destruction, inflammatory cells infiltration spinal cord injury. However, the role of long non‐coding RNAs (lncRNAs) induced pathogenesis MS EAE remains unknown. Herein, we found an IL‐17‐induced lncRNA AK018453 promoted TGF‐β receptor‐associated protein 1 (TRAP1) expression Smad‐dependent signaling mouse primary astrocytes. Knockdown significantly suppressed astrocytosis, attenuated phosphorylation Smad2/3, reduced NF‐κB p65 CBP/P300 binding to TRAP1 promoter, diminished production IL‐17‐treated knockdown lentiviral vector vivo dramatically inhibited inflammation prevented mice from demyelination during progression EAE. Together, these results suggest regulates IL‐17‐dependent response reactive potentially promotes via TRAP1/Smad pathway. Targeting this pathway may have therapeutic potential for intervening demyelinating diseases.

Immune responses are essential for the clearance of pathogens and repair injured tissues; however, if these not properly controlled, autoimmune diseases can occur. Autoimmune (ADs) a family disorders characterized by body's immune response being directed against its own tissues, with consequent chronic inflammation tissue damage. Despite enormous efforts to identify new drug targets develop therapies prevent ameliorate AD symptoms, no definitive solutions available today. Additionally, while substantial progress has been made in development some ADs, most treatments only symptoms and, general, ADs still incurable. Hundreds genetic loci have identified associated genome-wide association studies. However, whole list molecular factors that contribute pathogenesis is unknown. Noncoding (nc)RNAs, such as microRNAs, circular (circ)RNAs, long noncoding (lnc)RNAs, regulate gene expression at different levels various diseases, including serve potential well biomarkers disease progression therapy. An epidemiological association between multiple sclerosis (MS) and inflammatory bowel disease (IBD) is well established, but whether this reflects a shared genetic aetiology, consistent relationships exist MS the two predominant IBD subtypes, ulcerative colitis (UC) Crohn's (CD), remains unclear. Here, we use large-scale genome-wide study summary data to investigate architecture overall UC CD independently. We find significantly greater correlation than CD, identify three SNPs (rs13428812), (rs116555563) (rs13428812, rs9977672) in cross-trait meta-analyses. suggestive evidence for causal effect of on using Mendelian randomization, no or weak inconsistent MS. observe largely patterns tissue-specific heritability enrichment IBDs lung, spleen, whole blood small intestine, cell-type-specific CD4+ T cells lung CD8+ cytotoxic spleen. Central nervous system (CNS) disease is a general term for series of complex and diverse diseases, including Alzheimer’s (AD), Parkinson’s (PD), multiple sclerosis (MS), CNS tumors, stroke, epilepsy, amyotrophic lateral (ALS). Interneuron neuron-glia cells communicate with each other through their homeostatic microenvironment. Exosomes in the microenvironment have crucial impacts on interneuron by transferring contents, such as proteins, lipids, ncRNAs, constituting novel form cell-to-cell interaction communication. The pathogenesis of multiple sclerosis (MS) is influenced by the interaction genetic and epigenetic mechanisms. long noncoding RNA GAS5 acts as a competing endogenous for microRNA-137 involved in demyelination. We investigated association miR-137 expression their polymorphisms with MS susceptibility. One hundred eight patients 104 healthy controls were included. Expression analysis genotyping GAS5-rs2067079 miR-137-rs1625579 single nucleotide performed qPCR. Serum was upregulated, while serum downregulated compared controls. an excellent discriminator from (AUC = 0.97) negative independent predictor multivariate logistic analysis. positively correlated expanded disability status scale scores relapsing-remitting patients. rs2067079TT minor homozygote genotype associated increased risk, rs1625579G allele protective. rs1625579 showed age-specific effect, rs2067079 affected risk gender- manners. In patients, higher than other genotypes, did not differ between genotypes. Our results suggest biomarkers, marker severity. propose novel markers susceptibility, at least, possibly impacts crosstalk miR-137.
