The primary challenge in today's world of neuroscience is the search for new therapeutic possibilities neurodegenerative disease. Central to these disorders lies among other factors, aberrant folding, aggregation, and accumulation proteins, resulting formation toxic entities that contribute neuronal degeneration. This review concentrates on key proteins such as β-amyloid (Aβ), tau, α-synuclein, elucidating intricate molecular events underlying their misfolding aggregation. We critically evaluate mechanisms governing elimination misfolded shedding light potential strategies. specifically examine pathways endoplasmic reticulum (ER) unfolded protein response (UPR), chaperones, chaperone-mediated autophagy (CMA), intersecting signaling Keap1-Nrf2-ARE, along with connected through p62. Above all, we emphasize significance quality control mechanisms, encompassing interventions targeting regulation post-translational modifications, enhancement chaperones clearance. Additionally, focus current new, multi-target approaches. In conclusion, this systematically consolidates insights into emerging strategies predicated aggregates

The incidence and mortality of cancer are increasing, making it a leading cause death worldwide. Conventional treatments such as surgery, radiotherapy, chemotherapy face significant limitations due to therapeutic resistance. Autophagy, cellular self-degradation mechanism, plays crucial role in development, drug resistance, treatment. This review investigates the potential autophagy inhibition strategy for cancer. A systematic search was conducted on Embase, PubMed, Google Scholar databases from 1967 2024 identify studies inhibitors their mechanisms therapy. includes original articles utilizing vitro vivo experimental methods, literature reviews, clinical trials. Key terms used were "Autophagy", "Inhibitors", "Molecular mechanism", "Cancer therapy", "Clinical trials". Autophagy chloroquine (CQ) hydroxychloroquine (HCQ) have shown promise preclinical by inhibiting lysosomal acidification preventing autophagosome degradation. Other like wortmannin SAR405 target specific components pathway. Combining these with has demonstrated enhanced efficacy, cells more susceptible cytotoxic agents. Clinical trials involving CQ HCQ encouraging results, although further investigation is needed optimize use exhibits dual cancer, functioning both survival mechanism cell Targeting presents viable therapy, particularly when integrated existing treatments. However, complexity regulation side effects necessitate research develop precise context-specific approaches.

Cell death occurs continuously throughout individual development. By removing damaged or senescent cells, cell not only facilitates morphogenesis during the developmental process, but also contributes to maintaining homeostasis after birth. In addition, reduces spread of pathogens by eliminating infected cells. is categorized into two main forms: necrosis and programmed death. Programmed encompasses several types, including autophagy, pyroptosis, apoptosis, necroptosis, ferroptosis, PANoptosis. Autophagy, a mechanism that maintains cellular equilibrium via breakdown reutilization proteins organelles, implicated in regulating almost all forms pathological contexts. Notably, PANoptosis are directly classified as autophagy-mediated Therefore, autophagy presents therapeutic approach for treating diseases such inflammation tumors arise from abnormalities other This review focuses on crosstalk between modalities, providing new perspectives clinical interventions inflammatory neoplastic diseases.

Autophagy plays a complex impact role in tumor initiation and development. It serves as double-edged sword by supporting cell survival certain situations while also triggering autophagic death specific cellular contexts. Understanding the intricate functions mechanisms of autophagy tumors is crucial for guiding clinical approaches to cancer treatment. Recent studies highlight its significance various aspects biology. enables cells adapt survive unfavorable conditions recycling components. However, excessive or prolonged can lead self-destruction via process known death. Unraveling molecular underlying regulation development targeted therapeutic interventions. The study of chemicals extracted from natural sources should be encouraged due to the significant number cancer deaths each year and financial burden imposed by this disease on society. causes almost all cancers involve a combination lifestyle, environmental factors, genetic inherited factors. Modern medicine researchers are increasingly interested in traditional phytochemicals as they hold potential for new bioactive compounds with medical applications. Recent publications have provided evidence antitumor properties phytochemicals, key component Chinese medicine, thereby opening avenues their use modern medicine. Various studies demonstrated strong correlation between apoptosis autophagy, two critical mechanisms involved formation regulation, indicating diverse forms crosstalk them. Phytochemicals ability activate both pro-apoptotic pro-autophagic pathways. Therefore, understanding how influence relationship autophagy is crucial developing treatment strategy that targets these molecular mechanisms. This review aims explore anticancer effects, focusing role regulating which contributes uncontrolled tumor cell growth. Additionally, highlights limitations challenges current research methodologies while suggesting future field.

Stroke represents a main cause of death and permanent disability worldwide. The molecular mechanisms underlying cerebral injury in response to the ischemic insults are not completely understood. In this article, we summarize recent evidence regarding role autophagy pathogenesis stroke by reviewing data obtained murine models either transient or middle artery occlusion, stroke-prone spontaneously hypertensive rat. Few preliminary observational studies investigating subjects at high cerebrovascular risk cohorts patients were also reviewed. Autophagy plays dual neuronal vascular cells exerting both protective detrimental effects depending on its level, duration stress type involved. Protective exerts adaptive which reduce loss promote survival. On other hand, excessive activation leads cell increases brain injury. conclusion, reviewed suggests that proper manipulation may represent an interesting strategy prevent

Summary The NLRP3 inflammasome is a multiprotein complex that upon activation by the innate immune system drives broad inflammatory response. primary initial mediators of this response are pro‐IL‐1β and pro‐IL‐18, both which in an inactive form. Formation activates caspase‐1, cleaves pro‐IL‐18 triggers formation gasdermin D pores. Gasdermin pores allow for secretion active IL‐1β IL‐18 initiating organism‐wide can be beneficial to host; however, if inappropriately activated it lead significant pathology. While components known, precise details assembly less well defined conflicting. Here, we discuss several proposed pathways inflammasome. We examine role subcellular localization reciprocal regulation autophagy. focus on roles mitochondria mitophagy activating regulating Finally, detail impact pathologic responses development manifestations pulmonary disease.
