Penaeus vannamei hemocyanin (PvHMC) exhibits multifunctional roles in immunity, often mediated by various post-translational modifications. While linear ubiquitination catalyzed LUBAC mammals regulates immune signaling, its role crustacean immunity remains unclear. Here, we investigated the regulatory mechanism of PvHMC an E3 ligase PvLUBEL (a HOIP homolog), with emphasis on shrimp defending against diverse pathogens. We detected multiple tissues, including hemocytes and hepatopancreas. During Vibrio parahaemolyticus infection, expression level were suppressed, whereas infection white spot syndrome virus (WSSV) led to their upregulation. Structural analyses revealed that PvLUBEL, which shares a conserved RING-IBR-RING (RBR) domain mammalian HOIP, serves as catalytic subunit. Notably, inhibition via HOIPIN-1 covalent inhibitor) or RNA interference (RNAi) significantly reduced ubiquitination, thereby increasing pathogen proliferation decreasing host survival. The experimental myocardial infarction (MI) model originating from isoproterenol (ISO) is frequently preferred in research due to its similarity MI-induced damage humans. Beneficial effects of L-arginine (L-Arg), a semi-essential amino acid, cardiovascular diseases have been shown many studies. This study was carried out determine whether L-Arg pre-intervention has protective on heart tissue the MI model. 28 rats used were randomly divided into 4 equal groups: control, L-Arg, ISO, and L-Arg+ISO. Upon completion all applications, cardiac markers serum biochemical, histopathological, immunohistochemical examinations tissues performed. Cardiac markers, histopathological changes, oxidative stress, inflammation, apoptosis increased In addition, administration ISO deregulated OTULIN levels mitochondrial dynamics tissue. However, showed significant effect against changes ISO-induced MI. supplementation with cardioprotective may reduce risks possible pathophysiological processes

Abstract One of the adverse effects antineoplastic drug cisplatin (CS) is damage to testicular tissue. This study aimed examine potential therapeutic effect thymoquinone (TQ), a strong antioxidant, against caused by CS. In experiment, 28 rats were used, and randomly divided into four groups: control ( n = 7), CS + TQ 7). The experiment was called off after all treatments finished on day 15. Blood serum tissues utilized for biochemical, histological, immunohistochemical, mRNA expression, gene protein investigations. testosterone level decreased oxidative stress, histopathological damage, dysregulation in mitochondrial dynamics, inflammation apoptotic cells increased tissue due administration. supplementation showed anti-inflammatory, anti-apoptotic response CS-induced damage. addition, contributed reduction toxic regulating TNF-α/OTULIN/NF-κB pathway. may be strategy axis, inhibiting inflammation, apoptosis.

Abstract Linear ubiquitination is a distinct type of that involves attaching head-to-tail polyubiquitin chain to substrate protein. Early studies found linear ubiquitin chains are essential for the TNFα- and IL-1-mediated NF-κB signaling pathways. However, recent have discovered at least sixteen substrates, which exhibit broader activity than expected mediate many other pathways beyond signaling. Dysregulation in these has been linked types cancers, such as lymphoma, liver cancer, breast cancer.
