Abstract Lipid droplets (LDs) form inter‐organelle contacts with the endoplasmic reticulum (ER) that promote their biogenesis, while LD mitochondria enhance β‐oxidation of contained fatty acids. Viruses have been shown to take advantage lipid viral production, but it remains unclear whether they also modulate interactions between LDs and other organelles. Here, we showed coronavirus ORF6 protein targets is localized mitochondria‐LD ER‐LD contact sites, where regulates biogenesis lipolysis. Lipid droplets (LDs), once considered inert storage depots neutral lipids, are now recognized as dynamic organelles critical in lipid energy regulation. Myocardial ischemia/reperfusion injury (MIRI) is the leading cause of irreversible myocardial damage. A pivotal pathogenic factor (I/R)-induced cardiomyocyte ferroptosis, marked by iron overload and lipid peroxidation. However, impact droplet (LD) changes on I/R-induced ferroptosis unclear. In this study, an aggregation-induced emission probe, TPABTBP developed that used for imaging dynamic in LD during ferroptosis. exhibits excellent LD-specificity, superior capability monitoring lipophagy, remarkable photostability. Molecular dynamics (MD) simulation super-resolution fluorescence demonstrate specifically localized to phospholipid monolayer membrane LDs. Imaging LDs cardiomyocytes tissue model mice with MIRI reveals accumulation level increase early reperfusion stage (0-9 h) but decrease late (>24 via lipophagy. The inhibition breakdown significantly reduces peroxidation cardiomyocytes. Furthermore, it demonstrated chloroquine (CQ), FDA-approved autophagy modulator, can inhibit thereby attenuating mice. This study describes ischemia suggests a potential therapeutic target intervention.
