Polyphenols represent the most studied class of nutraceuticals that can be therapeutics for a large spectrum diseases, including cancer. In this study, we investigated first time antitumor activities polyphenol extract from Annurca apple (APE) in MDA-MB-231 triple negative breast cancer cells, and explored underlying mechanisms. APE selectively inhibited cell viability caused G2/M phase arrest associated with p27 phospho-cdc25C upregulation p21 downregulation. promoted reactive oxygen species (ROS) generation cells while it acted as antioxidant non-tumorigenic MCF10A cells. We demonstrated ROS represented primary step activity pretreatment N-acetylcysteine (NAC) prevented APE-induced arrest, apoptosis, autophagy. downregulated Dusp-1 induced significant increase JNK/c-Jun phosphorylation were both by NAC. Moreover, downregulation JNK its specific inhibitor SP600125 significantly diminished anticancer indicating sustained activation main mechanism death. also AKT several oncoproteins, such NF-kB, c-myc, β-catenin. light these results, may an attractive candidate drug development against

A moderate programmed physical has often been reported therapeutic both in adulthood aging, since capable promote fitness. Regular alleviates negative effects caused by free radicals offers many health benefits, including reduced all-cause mortality, sarcopenia skeletal muscle, chronic disease, premature death elderly people. However, performance also known induce oxidative stress, inflammation, muscle fatigue. Many efforts have carried out identify micronutrients natural compounds, nutraceuticals, prevent or attenuate exercise-induced stress inflammation. The aim this review discuss benefits deriving from constant activity intake compounds protect body stress. attention will focused mainly on three antioxidants, which are quercetin, resveratrol curcumin. Reactive oxygen species (ROS) are common by-products of normal aerobic cellular metabolism and play important physiological roles in intracellular cell signaling homeostasis. The human body is equipped with antioxidant systems to regulate the levels these free radicals maintain proper function. However, a condition known as oxidative stress (OS) occurs, when ROS overwhelm body’s ability readily detoxify them. Excessive amounts generated under OS conditions cause damage proteins, lipids, nucleic acids, severely compromising health contributing disease development, including cancer. Biomarkers can therefore be exploited tools assessment status humans. In present review, we discuss different approaches used for evaluation clinical samples. described methods limited their reflect on only partially, revealing need more integrative examining both pro- reactions higher sensitivity physiological/pathological alternations. Since the evolution of aerobic metabolism, reactive oxygen species (ROS) have represented significant challenges to diverse life forms. In recent decades, increasing knowledge has revealed a dual role for ROS in cell physiology, showing they serve as major source cellular damage while also functioning important signaling molecules various biological processes. Our understanding homeostasis and ROS-mediated pathways presumed that are ancient highly conserved mechanisms shared by most organisms. However, emerging evidence highlights complexity plasticity signaling, particularly animals evolved extreme environments. this review, we focus on generation, antioxidative systems main influenced ROS. addition, discuss ROS’s responsive transcription regulation how it may been shaped over course evolution.

Saponins are steroidal or triterpenoid glycoside that is distinguished by the soap-forming nature. Different saponins have been characterized and purified gaining attention in cancer chemotherapy. possess high structural diversity, which linked to anticancer activities. Several studies reported role of mechanism actions, including cell-cycle arrest, antioxidant activity, cellular invasion inhibition, induction apoptosis autophagy. Despite extensive research significant effects saponins, there currently no known FDA-approved saponin-based drugs. This can be attributed a number limitations, toxicities drug-likeness properties. Recent explored options such as combination therapy drug delivery systems ensure increased efficacy decreased toxicity saponin. review discusses current knowledge on different their activity mechanisms action, well promising within last two decades recommendations for future studies.
