Pancreatic ductal adenocarcinoma (PDAC) is one among the most lethal malignancies due to its aggressive behavior and resistance conventional therapies. Hypoxia significantly contributes cancer progression therapeutic of PDAC. microRNAs (miRNAs/miRs) have emerged as critical regulators various biological processes. miR-210 known "hypoxamir" prominent role in cellular responses hypoxia. In this study, we investigated multifaceted PDAC using knockout (KO) models elucidate functions under hypoxic conditions. Hypoxia-inducible factor-1α (HIF1-α), a key transcription factor activated response low oxygen levels, upregulates miR-210. maintains stem cell (CSC) phenotypes promotes epithelial-mesenchymal transition (EMT), which essential for tumor initiation, metastasis, resistance. Our findings demonstrate that regulates expression CSC markers, such CD24, CD44, CD133, EMT including E-cadherin, Vimentin, Snail. Specifically, depletion reversed marker levels Panc-1 MiaPaCa-2 cells. These regulatory actions facilitate more invasive treatment-resistant phenotype. Understanding network involving conditions may reveal new targets combating improving patient outcomes. data suggest regulator HIF1-α expression, EMT, stemness cells environments.
