The field of pharmacogenetics, the investigation influence one or more sequence variants on drug response phenotypes, is a special case pharmacogenomics, discipline that takes genome-wide approach. Massively parallel, next generation sequencing (NGS), has allowed pharmacogenetics to be subsumed by pharmacogenomics with respect identification associated responders and non-responders, optimal response, adverse reactions. A plethora rare common naturally-occurring GPCR must considered in context signals from across genome. Many fundamentals were established for G protein-coupled receptor (GPCR) genes because they are primary targets large number therapeutic drugs. Functional studies, demonstrating likely-pathogenic pathogenic variants, have been integral establishing models used

The management of orofacial pain to alleviate the quality life affected patients is becoming increasingly challenging for scientific research and healthcare professionals. From this perspective, in addition conventional therapies, new alternatives are being sought, looking at use both natural synthetic products. Cannabis sativa L. represents an interesting source bioactive compounds, including non-psychoactive cannabinoids, flavonoids, terpenes, many which effective improving intensity. Here, we aim analyze possible mechanisms action hemp-derived compounds responsible modulatory effects on pain-related pathways. ability these act multiple through a synergistic effect, reducing release inflammatory mediators regulating response endocannabinoid system, makes them agents alternative formulations be used pain.

Inflammatory bowel disease (IBD) is a general term used to describe group of chronic inflammatory conditions the gastrointestinal tract unknown etiology, including two primary forms: Crohn's (CD) and ulcerative colitis (UC). The endocannabinoid system (ECS) plays an important role in modulating many physiological processes intestinal homeostasis, modulation motility, visceral sensation, or immunomodulation inflammation IBD. It consists cannabinoid receptors (CB1 CB2), transporters for cellular uptake ligands, endogenous bioactive lipids (Anandamide 2-arachidonoylglycerol), enzymes responsible their synthesis degradation (fatty acid amide hydrolase monoacylglycerol lipase), manipulation which through agonists antagonists system, shows potential therapeutic ECS disease. This review summarizes components on inflammation, suggesting advantages cannabinoid-based therapies

Gut microbiota has implications in Central Nervous System (CNS) disorders. Our study systematically identified preclinical studies aimed to investigate the possible gut contribution neuropathy and neuropathic pain. The systematic review is reported accordance with PRISMA checklist guidelines outlined updated 2020. We included research articles reporting neuropathy-related behavioral evaluations and/or neurological scores coupled analysis performed by high-throughput technologies last ten years. Two investigators a search through 3 electronic bibliographic databases for full-text (PubMed, Scopus, EMBASE) three registries (Prospero, SyRF, bioRxiv), cross-references, linear searches. assessed methodological quality via CAMARADES appraised heterogeneous body of evidence narrative synthesis. In total, there were 19 eligible studies. most these reports showed significant changes setting conditions. major microbiome remodeling was fecal transplantation. Mechanistic proof gut-CNS communication achieved measuring inflammatory mediators, metabolic products, or neurotransmitters. As limitation, we found considerable heterogeneity across conclude that current understanding findings suggested an association between pain modifications. provides basis further targeting managing symptoms other neuroinflammation-based CNS protocol registered on international database Prospero under registration number (257628).

Abstract The discovery of the endocannabinoidome (eCBome) is evolving gradually with yet to be elucidated functional lipid mediators and receptors. diet modulates these bioactive lipids gut microbiome, both working in an entwined alliance. Mounting evidence suggests that, different ways a certain specialisation, signalling such as N -acylethanolamines (NAEs), 2-monoacylglycerols (2-MAGs), -acyl-amino acids (NAAs), along endocannabinoids (eCBs), can modulate physiological mechanisms underpinning appetite, food intake, macronutrient metabolism, pain sensation, blood pressure, mood, cognition, immunity. This knowledge has been primarily utilised pharmacology medicine develop many drugs targeting fine specific molecular pathways orchestrating eCB eCBome activity. Conversely, contribution dietary NAEs, 2-MAGs eCBs biological functions molecules little studied. Endocannabinoids (eCBs) and the expanded endocannabinoid system (ECS)-“endocannabinoidome”, consists of endogenous ligands, eCBs, their canonical non-canonical receptor subtypes, synthesizing metabolizing enzymes. This modulates a wide range body functions acts as retrograde signaling within central nervous (CNS) by inhibition classical transmitters, plays vital modulatory function on dopamine, major neurotransmitter in CNS. Dopamine is involved different behavioral processes contributes to brain disorders—including Parkinson’s disease, schizophrenia, drug addiction. After synthesis neuronal cytosol, dopamine packaged into synaptic vesicles until released extracellular signals. Calcium dependent activation results vesicular release interacts with systems. The ECS, among others, regulation interaction occurs either through direct or indirect mechanisms. cross-talk between ECS dopaminergic has important influence various dopamine-related neurobiological pathologic conditions investigating this might help identify therapeutic targets options disorders CNS associated dysregulation.
