The management of orofacial pain to alleviate the quality life affected patients is becoming increasingly challenging for scientific research and healthcare professionals. From this perspective, in addition conventional therapies, new alternatives are being sought, looking at use both natural synthetic products. Cannabis sativa L. represents an interesting source bioactive compounds, including non-psychoactive cannabinoids, flavonoids, terpenes, many which effective improving intensity. Here, we aim analyze possible mechanisms action hemp-derived compounds responsible modulatory effects on pain-related pathways. ability these act multiple through a synergistic effect, reducing release inflammatory mediators regulating response endocannabinoid system, makes them agents alternative formulations be used pain.

Gut microbiota has implications in Central Nervous System (CNS) disorders. Our study systematically identified preclinical studies aimed to investigate the possible gut contribution neuropathy and neuropathic pain. The systematic review is reported accordance with PRISMA checklist guidelines outlined updated 2020. We included research articles reporting neuropathy-related behavioral evaluations and/or neurological scores coupled analysis performed by high-throughput technologies last ten years. Two investigators a search through 3 electronic bibliographic databases for full-text (PubMed, Scopus, EMBASE) three registries (Prospero, SyRF, bioRxiv), cross-references, linear searches. assessed methodological quality via CAMARADES appraised heterogeneous body of evidence narrative synthesis. In total, there were 19 eligible studies. most these reports showed significant changes setting conditions. major microbiome remodeling was fecal transplantation. Mechanistic proof gut-CNS communication achieved measuring inflammatory mediators, metabolic products, or neurotransmitters. As limitation, we found considerable heterogeneity across conclude that current understanding findings suggested an association between pain modifications. provides basis further targeting managing symptoms other neuroinflammation-based CNS protocol registered on international database Prospero under registration number (257628).

Abstract The discovery of the endocannabinoidome (eCBome) is evolving gradually with yet to be elucidated functional lipid mediators and receptors. diet modulates these bioactive lipids gut microbiome, both working in an entwined alliance. Mounting evidence suggests that, different ways a certain specialisation, signalling such as N -acylethanolamines (NAEs), 2-monoacylglycerols (2-MAGs), -acyl-amino acids (NAAs), along endocannabinoids (eCBs), can modulate physiological mechanisms underpinning appetite, food intake, macronutrient metabolism, pain sensation, blood pressure, mood, cognition, immunity.
