Osteoarthritis (OA), a common and disabling degenerative joint disease, affects millions of people worldwide imposes considerable burden on patients society due to its high prevalence economic costs. The pathogenesis OA is closely related the progressive degradation articular cartilage accompany inflammation; however, itself cannot heal modulate inflammation lack nerves, blood vessels, lymph-vessels. Therefore, reliable effective methods treat remain highly desired. Local administration drugs or bioactive materials by intra-articular injection delivery system represents promising approach OA, especially considering prolonged retention, chondrocytes targeting, stimuli-responsive release achieve precision therapy. This article summarizes discusses advances in currently used systems (nanoparticle, hydrogel, liposome, microsphere) then focuses their applications treatment from perspective endogenous stimulus (redox reactions, pH, enzymes, temperature) exogenous (near-infrared, magnetic, ultrasound)-responsive release. Traumatic or degenerative defects of articular cartilage impair joint function, and the treatment damage remains a challenge. By mimicking extracellular matrix (ECM), exosome-seeded cryogels may enhance cell proliferation chondral repair. ECM-based were cryopolymerized with gelatin, chondroitin sulfate, various concentrations (0%, 0.3%, 0.5%, 1%) hyaluronic acid (HA), their water content, swelling ratio, porosity, mechanical properties, effects on viability evaluated. The regenerative bone marrow-derived mesenchymal stem (BM-MSC)-derived exosome (at concentration 106 particles/mL)-seeded 0.3% HA assessed in vitro surgically induced male New Zealand rabbit vivo. porosity significantly (p < 0.05) increased Young's modulus values decreased increasing concentrations. MTT assays revealed that developed biomaterials had no cytotoxic effects. optimal cryogel composition was HA, resulting favorable properties suitable strength. Exosomes alone promoted chondrocyte (with optical densities 58% 51% greater than control). facilitated ECM deposition sulfated glycosaminoglycan synthesis. Although we observed repair via Alcian blue staining both cryogel, layered arrangement chondrocytes superior to control when used. Osteoarthritis (OA) is a cartilage-degenerative joint disease. Mitophagy impacts articular cartilage damage. tRNA-derived small RNAs (tsRNAs) are one of the contents adipose mesenchymal stem cell (AMSC)-derived exosomes (AMSC-exos) and involved in disease progression. However, whether tsRNAs regulate mitophagy tsRNA-modified AMSC-exos improve OA via remain unclear. We performed RNA sequencing to identify OA-related tsRNAs, which were then loaded into AMSC-exos, exploring function mechanisms related vitro vivo. Overall, 53 differentially expressed (DEtsRNAs) identified between normal tissues, among 42 DEtsRNAs, including tsRNA-12391, downregulated group. Target genes tsRNA-12391 mainly participated mitophagy-related pathways such as Rap1 signaling pathway. Compared control group, mimics significantly promoted mitophagy, shown by upregulated expression PINK1 LC3 co-localization Mito-Tracker Green PINK1. Furthermore, effectively enhanced chondrogenesis chondrocytes, demonstrated elevated collagen II ACAN. with overexpression also facilitated Mechanistically, bound ATAD3A restricted ATAD31 from degrading PINK1, leading accumulation. reversed effects on chondrogenesis. interacting ATAD31; this may be novel therapeutic strategy for OA.
