Precise control of cellular signaling events during programmed cell death is crucial yet challenging for cancer therapy. The modulation signal transduction in cells holds promise but limited by the lack efficient, biocompatible, and spatiotemporally controllable approaches. Here we report a photodynamic strategy that modulates both apoptotic pyroptotic altering caspase-3 protein activity associated crosstalk. This employs mitochondria-targeting, near-infrared activatable probe (termed M-TOP) functions via type-I photochemical mechanism. M-TOP less dependent on oxygen more effective treating drug-resistant cells, even under hypoxic conditions. Our study shows higher doses induce caspase-3/gasdermin-E pathway, whereas lower lead to apoptosis. method across diverse gasdermin-E-expressing cells. Several studies have shown that berberine (BBR) is effective in protecting against myocardial ischemia‑reperfusion injury (MI/RI). However, the precise molecular mechanism remains elusive. The present study observed and safeguarding effect of BBR hypoxia/reoxygenation (H/R) H9c2 cells. pretreatment significantly improved decrease cell viability, P62 protein, Rho Family GTPase 3 (RhoE) ubiquinone subunit B8 ubiquinol‑cytochrome c reductase core protein U, Bcl‑2‑associated X protein/B‑cell lymphoma 2 ratio, glutathione (GSH) GSH/glutathione disulphide (GSSG) ratio induced by H/R, while reducing increase lactate dehydrogenase, microtubule‑associated 1 light caspase‑3 activity, reactive oxygen species, GSSG malonaldehyde caused H/R. Transmission electron microscopy LysoTracker Red DND‑99 staining results showed inhibited H/R‑induced excessive autophagy mediating RhoE. also mitochondrial permeability transition, maintained stability membrane potential, reduced apoptotic rate, increased level caspase‑3. protective effects were attenuated pAD/RhoE‑small hairpin RNA, rapamycin (an activator) compound C AMP‑activated kinase inhibitor). Atherosclerosis (AS), a significant contributor to cardiovascular disease (CVD), is steadily rising with the aging of global population. Pyroptosis and apoptosis, both caspase-mediated cell death mechanisms, play an essential role in occurrence progression AS. The human pineal gland primarily produces melatonin (MT), indoleamine hormone powerful anti-oxidative, anti-pyroptotic, anti-apoptotic properties. This study examined MT's anti-oxidative stress anti-pyroptotic effects on THP-1 macrophages treated nicotine.In vitro, were induced by 1 µM nicotine form pyroptosis model performed 30 mM MT for treatment. In vivo, ApoE-/- mice administered 0.1 mg/mL solution as drinking water, was intragastric administrated at 10 mg/kg/day. changes pyroptosis, oxidative detected.MT downregulated whose paralleled reduction reactive oxygen species (ROS) production, reversal sirtuin3 (SIRT3), Forkhead box O3 (FOXO3α) upregulation. also inhibited mainly caused interaction caspase-1 caspase-3 proteins. Bladder cancer (BC) is one of the most prevalent cancers worldwide. However, recurrence rate and five-year survival have not been significantly improved in advanced BC, new therapeutic strategies are urgently needed. The anticancer activity stellettin B (SP-2), a triterpene isolated from marine sponge Rhabdastrella sp., was evaluated with MTT assay as well PI Annexin V/7-AAD staining. Detailed mechanisms were elucidated through an NGS analysis, protein arrays, Western blotting. SP-2 suppressed viability BC cells without severe toxicity towards normal uroepithelial cells, it increased apoptosis activation caspase 3/8/9, PARP, γH2AX. phosphorylation FGFR3 its downstream targets downregulated by SP-2. Meanwhile, induced autophagy evidenced LC3-II formation p62 downregulation. inhibition using pharmacological inhibitors or ATG5-knockout protected RT-112 SP-2-induced cell suppression apoptosis. In addition, upregulation DAPK2 mRNA expression also contributed to cytotoxicity FGFR3-TACC3-knockout caused downregulation DAPK2, autophagy, conclusion, this first study demonstrating that exhibits potent anti-BC suppressing FGFR3-TACC3/Akt/mTOR pathway, which further activates novel autophagy/DAPK2/apoptosis signaling cascade.
