Background: Recently, multiple preclinical studies have reported the beneficial effect of berberine in treatment Alzheimer’s disease (AD). Nevertheless, neuroprotective effects and possible mechanisms against AD are not universally recognized. This study aimed to conduct a systematic review meta-analysis by integrating relevant animal assess potential on AD. Methods: We systematically searched PubMed, Embase, Scopus Web Science databases that models up 1 February 2023. The escape latency, times crossing platform, time spent target quadrant pro-oligomerized amyloid beta 42 (Aβ 1-42 ) were included as primary outcomes. secondary outcomes Tau-ps 204, 404, β-site APP cleaving enzyme (BACE1), precursor protein (APP), acetylcholine esterase (AChE), tumor necrosis factor ⍺ (TNF-α), interleukin 1β (IL-1β), IL-6, nitric oxide (NO), glial fibrillary acidic (GFAP), malonaldehyde (MDA), glutathione S-transferase (GST), (GSH), peroxidase (GPx), Beclin-1 neuronal apoptosis cells. was conducted using RevMan 5.4 STATA 15.1. SYRCLE’s risk bias tool used methodological quality. Results: Twenty-two 453 animals analysis. overall results showed significantly shortened latency ( p < 0.00001), increased platform 0.00001) decreased Aβ deposition 202 404 = 0.002), improved BACE1, APP, AChE, Beclin-1, cells, oxidative stress inflammation levels. Conclusion: Berberine may be promising drug for based evidence (especially when dose 5–260 mg/kg). these protective closely related anti-neuroinflammation, anti-oxidative stress, modulation autophagy, inhibition protection cholinergic system. However, limited quality existing research. Larger methodologically more rigorous research needed provide convincing evidence.

Deltamethrin (DM) is a highly effective and widely used pyrethroid pesticide. It an environmental factor affecting public occupational health exerts direct toxic effects on the central nervous system. As major target organs for neurotoxicity of DM, hippocampus cerebellum are critical to learning motor function. Pregnant Wistar rats were randomly divided into four groups gavaged at doses 0, 1, 4or 10 mg/kg/d DM from gestational day (GD) 0 postnatal (PN) 21. The PC12 cells selected further verify regulatory mechanisms neurodevelopmental injury. We found that maternal exposure caused learning, memory dysfunction in male offspring. Maternal induced decrease density hippocampal dendritic spines offspring through reduced expression M1 mAchRs, which turn mediated AKT/mTOR signaling pathway, contributing inhibition dynamic changes GluA1. Meanwhile, inhibited BDNF/TrkB thereby reducing phosphorylation stathmin impairing cerebellar purkinje cell dendrite growth development. Taken together, during pregnancy lactation could impair neurodevelopment

Several reports have stated the neuroprotective and learning/memory effects of Tachyspermum ammi seed extract (TASE) its principal component thymol; however, little is known about underlying molecular mechanisms neurogenesis potential. This study aimed to provide insights into TASE a thymol-mediated multifactorial therapeutic approach in scopolamine-induced Alzheimer's disease (AD) mouse model. thymol supplementation significantly reduced oxidative stress markers such as brain glutathione, hydrogen peroxide, malondialdehyde whole homogenates. Tumor necrosis factor-alpha was downregulated, whereas elevation brain-derived neurotrophic factor phospho-glycogen synthase kinase-3 beta (serine 9) enhanced learning memory TASE- thymol-treated groups. A significant reduction accumulation Aβ 1-42 peptides observed brains mice. Furthermore, promoted adult neurogenesis, with increased doublecortin positive neurons subgranular polymorphic zones dentate gyrus treated-mice. Collectively, could potentially act natural agents for treatment neurodegenerative disorders, AD.

Aging is associated with impairments in learning, memory, and cognitive flexibility, as well a gradual decline hippocampal neurogenesis. We investigated the performance of 6-and 14-month-old mice (considered mature adult late middle age, respectively) learning memory tasks based on Morris water maze (MWM) determined their levels preceding current While both age groups successfully performed spatial version MWM (sMWM), older were less efficient compared to younger when presented modified versions that required reassessment previously acquired experience. This was detected reversal (rMWM) particularly evident context discrimination (cdMWM), novel task integrating various distal cues, local altered contexts adjusting used search strategies. Older impaired several metrics characterize rMWM cdMWM, however, they showed improvement narrowed gap after additional training. Furthermore, we analyzed adult-born immature neurons dentate gyrus found significant correlation between neurogenesis individual demanding flexibility. These results provide detailed description age-related changes underscore importance supporting

Abstract The neurosphere assay is the gold standard for determining proliferative and differentiation potential of neural progenitor cells (NPCs) in neurogenesis studies 1–3 . While several vitro assays have been developed to model process neurogenesis, they predominantly used embryonic early postnatal NPCs derived from dentate gyrus (DG). A limitation these approaches that do not provide insight into adult-born NPCs, which are modeled affect hippocampal function diseases later life. Here, we show a novel free-floating culture system using isolated DG mature adult aged mice. protocol outlines detailed steps on isolation, propagation, maintenance neurospheres (>12 months old) mouse brain how differentiate cultured neurons astrocytes. Culturing provides an important (1) investigate cellular molecular properties this unique cell population (2) expand understanding plasticity aging brain. This requires ∼2 hours complete dissection, dissociation plating, while neuronal astrocytic lineages takes 9 days. By focusing obtained animals at ages facilitates investigation biological questions related development generated throughout

Abstract A microdeletion of approximately 2.4 Mb at the 8p23 terminal region has been identified in a Taiwanese autistic boy. Among products transcribed/translated from genes mapped this region, reduction DLGAP2, postsynaptic scaffold protein, might be involved pathogenesis autism spectrum disorder (ASD). DLGAP2 protein was detected hippocampus yet abolished homozygous Dlgap2 knockout (Dlgap2 KO) mice. In study, we characterized hippocampal phenotypes mutant KO mice exhibited impaired spatial memory, indicating poor function absence DLGAP2. Aberrant expressions proteins, including PSD95, SHANK3, HOMER1, GluN2A, GluR2, mGluR1, mGluR5, βCAMKII, ERK1/2, ARC, BDNF, were noticed Further, spine density increased mice, while ratio mushroom-type spines decreased. We also observed thinner thickness ultrastructural level. These structural changes found linked to hippocampus-related cognitive functions such as memory. Mice with deficiency, showing signs intellectual disability, common co-occurring condition patients ASD, could promising animal model which may advance our understanding ASD.
