Marine polyphenols, including eckol(EK), dieckol(DK), and 8,8'-bieckol(BK), have attracted attention as bioactive ingredients for preventing Alzheimer's disease (AD). Since AD is a multifactorial disorder, the present study aims to provide an unbiased elucidation of unexplored targets mechanisms systematic prediction effective preventive combinations marine polyphenols. Based on omics data between each compound AD, protein-protein interaction (PPI) network was constructed predict potential hub genes. Gene Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) pathway analyses were performed further biological insights. In PPI top 10 genes, AKT1, SRC, EGFR, ESR1 common EK BK, whereas PTGS2 target DK BK. GO KEGG analysis revealed that overlapped genes mainly enriched in EGFR tyrosine kinase inhibitor resistance, MAPK pathway, Rap1 Ras pathways. Finally, docking validation showed stable binding polyphenols their gene via lowest energy multiple interactions. The results expanded novel also provided system-level insight into molecular against AD.

Psychiatric and neurodegenerative disorders are amongst the most prevalent debilitating diseases, but current treatments either have low success rates, greatly due to permeability of blood–brain barrier, and/or connected severe side effects. Hence, new strategies extremely important, here is where liposome-derived nanosystems come in. Niosomes, transfersomes, ethosomes nanometric vesicular structures that allow drug encapsulation, protecting them from degradation, increasing their solubility, permeability, brain targeting, bioavailability. This review highlighted great potential these for treatment Alzheimer’s disease, Parkinson’s schizophrenia, bipolar disorder, anxiety, depression. Studies regarding encapsulation synthetic natural-derived molecules in systems, intravenous, oral, transdermal, or intranasal administration, led an increased bioavailability when compared conventional pharmaceutical forms. Moreover, developed formulations proved neuroprotective, anti-inflammatory, antioxidant effects, including neurotransmitter level restoration oxidative status improvement, improved locomotor activity enhancement recognition working memories animal models. albeit being relatively technologies, niosomes, already proven increase psychoactive drugs, leading effectiveness decreased showing promise as future therapeutics.

Alzheimer disease (AD) is the most prevalent form of dementia among elderly people. Owing to its varied and multicausal etiopathology, intervention strategies have been highly diverse. Despite ongoing advances in field, efficient therapies mitigate AD symptoms or delay their progression are still limited scope. Neuroplasticity, broad terms ability brain modify structure response external stimulation damage, has received growing attention as a possible therapeutic target, since disruption plastic mechanisms appear correlate with various forms cognitive impairment present patients. Several pre-clinical clinical studies attempted enhance neuroplasticity via different mechanisms, for example, regulating glucose lipid metabolism, targeting activity neurotransmitter systems, addressing neuroinflammation. In this review, we first describe several structural functional aspects neuroplasticity. We then focus on current status pharmacological approaches stemming from trials neuroplastic This followed by an analysis analogous interventions animal models, according action.

To address the challenges posed by biofilm presence and achieve a substantial reduction in bacterial load within root canals during endodontic treatment, various irrigants, including nanoparticle suspensions, have been recommended. Berberine (BBR), natural alkaloid derived from plants, has demonstrated potential applications dentistry treatments due to its prominent antimicrobial, anti-inflammatory, antioxidant properties. This study aimed produce characterize novel polymeric of poly (lactic-co-glycolic acid) (PLGA) loaded with berberine evaluate antimicrobial activity against relevant pathogens, Enterococcus faecalis, Candida albicans. Additionally, cytocompatibility using gingival fibroblasts was assessed. The prepared nanoprecipitation method. Physicochemical characterization revealed spheric nanoparticles around 140 nm ca, −6 mV surface charge, which unaffected BBR. successfully incorporated at an encapsulation efficiency 77% designed were stable upon 20 weeks storage 4 °C 25 °C. Free BBR reduced planktonic growth ≥125 μg/mL. Upon incorporation into PLGA nanoparticles, μg/mL [BBR]-loaded lead significant reduction, after 1 h contact, both bacteria yeast. Sessile cells biofilms also considered. At 30 40 μg/mL, viability sessile bacteria, 24 exposure. cytotoxicity BBR-loaded oral negligible. berberine-loaded hold as promising supplementary approach treatment infections.

Insights into role retinoic acid receptor-α, small nucleolar RNA host gene 1, and brain-derived neurotrophic factor (RARα-Snhg1-Bdnf) network can offer significant advancements in minimizing these effects. study targets exploration RARα Snhg1 regulatory network's Bdnf expression realm propofol-induced neurotoxicity. Harnessing Gene Expression Omnibus (GEO) database utilizing JASPAR RNA-Protein Interaction Prediction (RPISeq) for projections, embarks in-depth analysis employing both vitro vivo models. findings draw a clear link between neurotoxicity amplification RAR signaling pathways, impacting hippocampal development apoptosis leading increased decreased Bdnf. Propofol is inferred accentuate by heightening interactions, culminating suppression.

Inflammatory bowel diseases (IBDs) are characterized by chronic relapsing intestinal inflammation that causes digestive system dysfunction. For years, researchers have been working to find more effective and safer therapeutic strategies treat these diseases. Silibinin (SIL), a flavonoid compound extracted from the seeds of milk thistle plants, possesses multiple biological activities is traditionally applied liver SIL also widely used in treatment variety inflammatory attributed its excellent antioxidant anti-inflammatory effects. However, efficacy against IBDs mechanisms remain unclear. In this study, using Drosophila melanogaster as model organism, we found can effectively relieve caused dextran sulfate sodium (DSS). Our results suggested supplementation inhibit overproliferation stem cells (ISCs) induced DSS, protect barrier function, acid-base balance, excretion reduce reactive oxygen species (ROS) levels stress, extend lifespan Drosophila. Furthermore, our study demonstrated ameliorates via modulating c-Jun N-terminal kinase (JNK) signaling pathway research aims provide new insight into IBDs.

Background and aim Pathological changes in the central nervous system (CNS) begin before clinical symptoms of Alzheimer’s Disease (AD) manifest, with hippocampus being one first affected structures. Current treatments fail to alter AD progression. Traditional Chinese medicine (TCM) has shown potential improving pathology through multi-target mechanisms. This study investigates pathological hippocampal tissue explores TCM active components that may alleviate these changes. Methods GSE5281 GSE173955 datasets were downloaded from GEO normalized identify differentially expressed genes (DEGs). Key functional modules hub analyzed using Cytoscape R. Active identified literature Pharmacopoeia People’s Republic China. Enrichment analyses performed on target overlapping DEGs. Result From datasets, 76 upregulated 363 downregulated identified. Hub included SLAMF, CD34, ELN (upregulated) ATP5F1B, VDAC1, VDAC2, HSPA8, ATP5F1C, PDHA1, UBB, SNCA, YWHAZ, PGK1 (downregulated). Literature review 33 23 herbal medicines. Target gene enrichment analysis for six components: dihydroartemisinin, berberine, naringenin, calycosin, echinacoside, icariside II. Conclusion Mitochondrial synaptic vesicle dysfunction pathways enriched genes.
