One of the major obstacles confronting formulation a mechanistic understanding for Alzheimer’s disease (AD) is its immense complexity—a complexity that traverses full structural and phenomenological spectrum, including molecular, macromolecular, cellular, neurological behavioural processes. This reflected by equally complex diversity risk factors associated with AD. However, more than merely mirroring complexity, also provide fundamental insights into aetiology pathogenesis AD as neurodegenerative disorder since they are central to initiation subsequent propagation. Based on systematic literature assessment, this review identified 30 then extended analysis further identify neuroinflammation unifying mechanism present in all factors. Although other mechanisms (e.g., vasculopathy, proteopathy) were multiple factors, dysfunction neuroimmune–neuroinflammation axis was uniquely Though nature neuroinflammatory involvement varied, activation microglia release pro-inflammatory cytokines common pathway shared observation provides evidence importance immunopathic aetiopathogenesis

Abstract: We present the case of a 58-year-old male patient who presented with pruritic skin plaques and papules on scalp, face, back, back hands for over year. The symptoms worsened upon exposure to sunlight improved cloudy days. Despite previous attempts at treatment glucocorticoid ointment antihistamine drugs, experienced progressive aggravation symptoms. Physical examination revealed hypertrophic infiltrating nodules, significant scratches local exudation. Skin biopsy confirmed diagnosis sun-induced dermatosis. was initiated tofacitinib, an oral Janus Kinase inhibitor, along halometasone ointment, ebastine tablets, strict sun protection. Over course four revisits spanning months, improvement in symptoms, rash almost disappearing pruritus subsiding. well tolerated no adverse effects were observed. Follow-up six months post-treatment showed recurrence This highlights efficacy tofacitinib managing suggests it as potential therapeutic option similar cases. Background and Objective: Alzheimer's disease (AD) is a progressive neurodegenerative disorder, that is, characterized by cognitive decline. To date, there are no effective treatments for AD. Therefore, the objective of this study was to map new perspectives on effects pharmacological treatment function overall psychological state in patients with Methods: Two independent researchers searched randomized clinical trials (RCTs) exploring approaches related cognition adults from 2018 2023 PubMed, Web Science, Scopus, Cochrane Library databases. A total 17 RCTs were included review. Results: The results show recent years, drugs have been tested disease, including masitinib, methylphenidate, levetiracetam, Jiannao Yizhi, Huannao Yicong formulas. Most studies conducted populations mild moderate disease. Conclusion: Although some found suggested improvement function, scarcity available highlights need further research area. Alzheimer's disease (AD), the major form of neurodegenerative diseases that can severely impede normal cognitive function, makes it one most common fatal diseases. There are currently over 50 million AD patients worldwide. The neuropathology is perplexing and there a scarcity disease-modifying treatments. Currently, early diagnosis has been made possible with discovery biological markers associated pathology, providing strong support for improvement status. search inhibitors from dietary supplements (DSs) become hot topic. Especially widespread use DSs, DSs containing polyphenols, alkaloids, terpenes, polysaccharides other bioactive components prevent by reducing Aβ deposition, inhibiting tau protein hyperphosphorylation, reconstructing synaptic dysfunction, weakening cholinesterase activity, regulating mitochondrial oxidative stress, neuronal inflammation apoptosis. This review summarizes anti-AD effects main their constituents, as well potential molecular mechanisms covers 2017 to 2023. Additionally, we discussed opportunities challenges faced in process prevention treatment, aiming further provide new perspectives functional food development.
