The potential of natural phytochemicals in mitigating depression has been supported by substantial evidence. This study evaluated a total 88 with antidepressant properties targeting serotonin (5-HT) receptors (5-HT1A, 5-HT4, and 5-HT7) using molecular docking, ADMET (Absorption, Distribution, Metabolism, Excretion, Toxicity) analysis, internal coordinates normal mode analysis (NMA), dynamics simulation (MDS), free energy calculation. Five compounds (Genistein, Kaempferol, Daidzein, Peonidin, glycitein) exhibited favorable pharmacokinetic improved binding scores, indicating their as effective antidepressants. Redocking superimposition 5-HT cocrystal structures validated these findings. Furthermore, NMA, MDS, calculations confirmed the stability deformability ligand-receptor complexes, suggesting that can effectively interact to modulate depressive symptoms. These powerful phytochemicals, abundantly found soybeans, fruits, vegetables, herbs, represent promising avenue for developing treatments depression. Further vitro vivo studies are warranted explore efficacy alleviating stress through interactions receptors.

The tryptophan-kynurenine (KYN) pathway has long been recognized for its essential role in generating metabolites that influence various physiological processes. Traditionally, these have categorized into distinct, often opposing groups, such as pro-oxidant versus antioxidant, excitotoxic/neurotoxic neuroprotective. This dichotomous framework shaped much of the research on conditions like neurodegenerative and neuropsychiatric disorders, well cancer, where metabolic imbalances are a key feature. effects significantly influenced by factors, including concentration particular cellular milieu which they generated. Citrus species have a history of use in treating neurological conditions like insomnia, anxiety, and nervousness, though these claims lack exhaustive validation. This study evaluated the anxiolytic activity paradisi Macfayden var. redblush leaves, aiming to isolate characterise bioactive constituent. Four leaf extracts (petroleum ether (60–80 °C), chloroform, methanol aqueous) were for using elevated plus maze (EPM) mice. The extract showed was subjected bioactivity guided isolation solvent partitioning, column chromatography preparative HPLC compound, CP-1, characterised as rhoifolin (based on spectroscopic analysis). Rhoifolin, at 0.705% w/w confirmed compound through additional tests (light-dark box social interaction tests) holds promise potential treatment anxiety disorders.

Parkinson's disease (PD) is one of the most common neurodegenerative diseases. Constipation a prodromal symptom PD. It important to investigate pathogenesis constipation symptoms in Rotenone has been successfully used establish PD animal models. However, specific mechanism rotenone-induced not well understood. In this work, we found that appeared earlier than motor impairment mice gavaged with low dose rotenone (30 mg/kg·BW). only caused loss dopaminergic neurons and accumulation α-synuclein, but also significantly reduced serum 5-HT levels 5-HTR4 striatum colon. Background/Objectives: Globally, there has been an increase in the prevalence of anxiety and depression among university students, medical students are no exception. Medical especially susceptible to these mental health challenges, primarily due multifaceted stressors, which can significantly impact their academic achievements future career. There is a pressing need for comprehensive research that not only investigates but also explores strategies developing effective interventions support systems enhance well-being students. Therefore, this study aimed identify severity at South Africa, evaluating association socio-demographic, student, clinical variables with total general disorder (GAD-7) patient questionnaire (PHQ-9) scores. Methods: A survey-based quantitative cross-sectional was conducted 208 undergraduate African university. Participants who provided written consent completed GAD-7 PHQ-9 questionnaires together variable information. The relationship between scores determined using Mann–Whitney U test Kruskal–Wallis H test. Correlation analysis used establish threshold statistical significance set p ≤ 0.05. Results: More than half participants were female (n = 130; 62.5%), single 123; 59.1%), belonged Pedi ethnic group. majority Christian 183; 88.0%), received bursary had rural background 155; 74.5%). ABSTRACT Chronic stress increases susceptibility to anxiety and depression disorders, recurrent common psychiatric conditions. Current antidepressant medications have varying degrees of efficacy often multiple side effects limiting treatment adherence. Physical exercise has beneficial on stress‐related mental disorders. However, the underlying mechanisms are unclear. Dentate gyrus granule cells (GCs) excitability may mediate resilience. Here, we expose young adult C57Bl6 mice chronic restraint (CRS) for 14 days followed by 30 running treatment. Behavioural evaluation before after showed that behavioural alterations elicited CRS were mitigated running. Next, evaluated serotonergic modulation GC excitability, as a potential mechanism running‐induced Electrophysiological recordings indicate alters excitability. Utilising (S)‐WAY 100135 Tropisetron, antagonists 5‐HT 1A 3 receptors respectively, show recovers receptor activity lost CRS. Additionally, promotes indirect GCs through activation. Thus, be targets
