This review provides comprehensive synthesis of interplay between sarcopenia cardiogeriatrics, emphasizing shared mechanisms such chronic low-grade inflammation (inflammaging), hormonal dysregulation, oxidative stress, physical inactivity. Despite advancements diagnostic frameworks, EWGSOP2 AWGS definitions, variability criteria assessment methods continues to challenge standardization. Key tools include dual-energy X-ray absorptiometry (DXA) bioimpedance analysis (BIA) for alongside functional measures grip strength gait speed. The highlights bidirectional relationship conditions heart failure, aortic stenosis, atherosclerotic disease, which exacerbate each other through complex pathophysiological mechanisms. The triad of vascular impairment, muscle atrophy, and cognitive decline represents critical age-related conditions that significantly impact health. Vascular impairment disrupts blood flow, precipitating the mass reduction seen in sarcopenia neuronal function characteristic neurodegeneration. Our limited understanding intricate relationships within this hinders accurate diagnosis effective treatment strategies. This review analyzes interrelated mechanisms contribute to these conditions, with a specific focus on oxidative stress, chronic inflammation, impaired nutrient delivery. aim is understand common pathways involved suggest comprehensive therapeutic approaches. dysfunctions hinder circulation transportation nutrients, resulting characterized by atrophy weakness. dysfunction have negative physical quality life. Neurodegenerative diseases exhibit comparable pathophysiological affect motor functions. Preventive approaches encompass lifestyle adjustments, addressing integrated therapies improving muscular well-being. Better links can refine strategies yield better patient outcomes. study emphasizes complex interplay between dysfunction, degeneration, decline, highlighting necessity for multidisciplinary Advances domain promise improved diagnostic accuracy, more options, enhanced preventive measures, all contributing higher life elderly population.

Cell therapy based on mesenchymal stem cells (MSCs) alleviate muscle atrophy caused by diabetes and aging; however, the impact of human umbilical cord following nerve injury underlying mechanisms remain unclear. In this study, we evaluated therapeutic efficacy MSCs (hucMSCs) hucMSC-derived exosomes (hucMSC-EXOs) for identified molecular mechanisms. Sciatic crush in rats induction myotubes L6 were used to determine ameliorating effect hucMSCs hucMSC-EXOs atrophy. Q-PCR Western blot analyses measure expression muscle-specific ubiquitin ligases Fbxo32 (Atrogin1, MAFbx) Trim63 (MuRF-1). Dual-luciferase reporter gene experiments conducted validate direct binding miRNAs their target genes. Local injection mitigated rat gastrocnemius sciatic injury. vitro, alleviated myotubes. Mechanistic analysis indicated upregulation miR-23b-3p levels treatment. MiR-23b-3p significantly inhibited its genes, Trim63, suppressed myotube Notably, an inhibitor reversed inhibitory vitro. These results suggest that secreted contributes mechanism inhibiting ubiquitination Trim63.

Abstract Background Gastric cancer remains a major health concern worldwide, with locally advanced gastric (LAGC) posing significant challenges due to frequent lymph node metastasis and poor prognosis. While curative gastrectomy D2 dissection is the standard treatment, sarcopenia perioperative skeletal muscle loss (SML) have emerged as critical prognostic factors. Methods We retrospectively analyzed 198 LAGC patients who underwent gastrectomy. Preoperative postoperative were assessed via computed tomography (CT)-derived index (SMI) at L3 level. SML was defined based on sex-specific thresholds of SMI changes (%/30 days). Prognostic significance for overall survival (OS) disease-free (DFS) evaluated using Kaplan-Meier Cox regression analyses. Results The prevalence increased from 23.7% preoperatively 33.3% postoperatively. Patients showed worse OS DFS compared non-SML ( P < 0.05). also associated higher incidence Clavien-Dindo grade ≥ 3a complications Multivariate analysis identified preoperative (HR = 2.332, 0.001), 3.189, 0.011), 11.231, 0.002) independent risk factors OS. Adjuvant chemotherapy significantly improved both 0.532, 0.015) 0.587, 0.041). Conclusions Both may negatively impact prognosis in patients, suggesting that evaluation mass help identify high-risk surgical candidates targeted interventions.
