Breast cancer (BC) is a heterogenous disease, with prognosis and treatment options depending on Estrogen, Progesterone receptor, Human Epidermal Growth Factor Receptor-2 (HER-2) status. HER-2 negative, endocrine-independent BC presents significant clinical challenge limited options. To date, promising strategies like immune checkpoint inhibitors have not yielded breakthroughs in patient prognosis. Despite being considered archaic, agents derived from natural sources, mainly plants, remain backbone of current treatment. In this context, we critically analyze novel naturally-derived drug candidates, elucidate their intricate mechanisms action, evaluate pre-clinical vitro vivo activity negative BC. Since research success often does directly correlate approval, focus ongoing trials to uncover trends. Abstract Triple-negative breast cancer (TNBC) is a subtype of human with one the worst prognoses, no targeted therapeutic strategies currently available. Regulated cell death (RCD), also known as programmed (PCD), has been widely reported to have numerous links progression and therapy many types cancer. Of note, RCD can be divided into different subroutines, including autophagy-dependent death, apoptosis, mitotic catastrophe, necroptosis, ferroptosis, pyroptosis anoikis. More recently, targeting subroutines small-molecule compounds emerging promising strategy, which rapidly progressed in treatment TNBC. Therefore, this review, we focus on summarizing molecular mechanisms above-mentioned seven major related TNBC latest progress subroutines. Moreover, further discuss combined drug (e.g., narciclasine) or more drugs torin-1 chloroquine) achieve potential by regulating importantly, demonstrate several ONC201 NCT03733119) clinical trials. Breast cancer is a leadingcause of cancer-related deaths in women globally, with triple-negative breast (TNBC) being an aggressive subtype that lacks targeted therapies and associated poor prognosis. Polyphenols, naturally occurring compounds plants, have been investigated as potential therapeutic strategy for TNBC. This review provides overview the anticancer effects polyphenols TNBC their mechanisms action. Several polyphenols, including resveratrol, quercetin, kaempferol, genistein, epigallocatechin-3-gallate, apigenin, fisetin, hesperetin luteolin, shown to inhibit cell proliferation, induce cycle arrest, promote apoptosis, suppress migration/invasion preclinical models. Abstract Resveratrol, a natural polyphenol in various plants, has gained significant attention for its potential health‐promoting properties. It been demonstrated, after reviewing clinical and vitro studies, that resveratrol possesses potent antioxidant potential. Resveratrol demonstrates cellular component protection by directly neutralizing free radicals (FRs) enhancing the expression of enzymes, thereby mitigating oxidative damage to proteins, lipids, nucleic acids. Clinical trials have shown promising results, indicating supplementation can enhance defenses reduce markers populations. In addition effects, exhibits anti‐inflammatory modulate key inflammatory pathways, such as nuclear factor‐kappa B (NF‐κB) mitogen‐activated protein kinases (MAPKs), suppressing production pro‐inflammatory cytokines chemokines. Extensive investigation has been conducted on plant-based resources for their pharmacological usefulness, including various cancer types. The scope of this review is wider than several studies with a particular focus breast cancer, which an international health concern while studying sources flavonoids, carotenoids, polyphenols, saponins, phenolic compounds, terpenoids, and glycosides apart from focusing nursing. Important findings prior are synthesized to explore these compounds' sources, mechanisms action, complementary synergistic effects, associated side effects.
