Cancer metastasis still accounts for up to 90% of cancer-related deaths, but the molecular mechanism is unclear. Several chemokines and their receptors mediate tumor cell metastasis, particularly through long-term effects that regulate angiogenesis, proliferation apoptosis. Among them, CXC chemokine receptor 4 (CXCR4) has been shown play a pivotal role in cancer interaction with ligand (CXCL12), also known as stromal cell-derived factor 1α (SDF-1α). The CXCR4 promoter region well characterized, its expression controlled by various transcriptional factors, including NF-κB, HIF-1α, so forth. Isoorientin (ISO) 3', 4', 5, 7-tetrahydroxy-6-C-glucopyranosyl flavone. ISO reported exhibit anti-oxidant, anti-cancer, anti-inflammatory properties. However, anti-metastatic effect following downregulation unknown, underlying antitumor activity yet be elucidated. In our present study, we showed inhibited NF-κB regulation breast colon cells. We have demonstrated inhibits variety Furthermore, found regulated inhibition transcription process. Inhibition reduced invasion cells CXCL12. conclusion, results suggest novel inhibitor key molecule contributing activity.

Exosomes, as a class of small extracellular vesicles closely related to the biological behavior various types tumors, are currently attracting research attention in cancer diagnosis and treatment. Regarding diagnosis, stability their membrane structure wide distribution body fluids render exosomes promising biomarkers. It is expected that exosome-based liquid biopsy will become an important tool for tumor future. For treatment, exosomes, “golden communicators” between cells, can be designed deliver different drugs, aiming achieve low-toxicity low-immunogenicity targeted delivery. Signaling pathways exosome contents also used safer more effective immunotherapy against tumors. Exosomes derived from range sources, exhibit characteristics well clinical application advantages therapies. In this review, we analyzed main sources have great potential broad prospects therapy. Moreover, compared therapeutic advantages, providing new ideas exosomes.

Background: Brain cancers represent a formidable oncological challenge characterized by their aggressive nature and resistance to conventional therapeutic interventions. The tumor microenvironment has emerged as critical determinant of progression treatment efficacy. Within this complex ecosystem, microglia macrophages play fundamental roles, forming intricate networks with peripheral immune cell populations, particularly T cells. precise mechanisms underlying microglial interactions cells contributions immunosuppression remain incompletely understood. Methods: This review comprehensively examines the cellular dialogue between in two prominent brain malignancies: primary glioblastoma secondary metastases. Results: Through comprehensive current scientific literature, we explore nuanced through which microglial-T modulate growth responses. Conclusions: Our analysis seeks unravel communication pathways that potentially underpin progression, ultimate goal illuminating novel strategies for cancer intervention.

Cancer-associated fibroblasts (CAFs) play a crucial role in the tumor microenvironment by promoting growth, immune evasion, and metastasis. Recently, drug delivery systems targeting CAFs have emerged as promising long-term effective approach to cancer treatment. Advances nanotechnology, particular, led development of nanomedicine designed specifically target CAFs, offering new possibilities for precise personalized therapies. This article reviews recent progress using nanocarriers that CAFs. Additionally, we explore potential combining multiple therapies, such chemotherapy immunotherapy, with enhance efficacy overcome resistance. Although many preclinical studies show promise, clinical application still faces considerable challenges, especially terms penetration large-scale production. Therefore, this review aims provide fresh perspective on CAF-targeted highlight future research directions applications.

Abstract Malignant tumours of the digestive system cover a wide range diseases that affect health people to large extent. Angiogenesis is indispensable in development, and metastasis tumours, mainly two ways: occupation or formation. Vessels can provide nutrients, oxygen, growth factors for encourage metastasis, so cancer progression depends on simultaneous angiogenesis. Recently, exosomes have been proven participate angiogenesis tumours. They influence by binding tyrosine kinase receptors (VEGFR)-1, VEGFR-2, VEGFR-3 with different affinities, regulating Yap-VEGF pathway, Akt pathway other signaling pathway. Additionally, are potential therapeutic vectors deliver many types cargoes cells. In this review, we summarize roles highlight clinical application prospects, directly used as targers delivery vehicles, antiangiogenic therapy.

Glioma is a common type of malignant tumor with high global incidence and limited therapeutic options. Buthus martensii Karsch (BmK) Scorpion listed as medicinal functional food material for centuries to relieve neurological diseases including glioma in traditional Chinese medicine. In this study, network pharmacology firstly revealed that BmK may exert anti-tumor effect via regulating oncogenic signals like EGFR SRC. The anti-neoplastic safety were further validated on two vivo models, subcutaneous xenograft orthotopic transplanted C57BL/6J mice. Data from mRNA miRNA transcriptomes showed exerted anti-glioma well immune regulatory effects. Flow cytometry immunofluorescence analysis demonstrated improved CD4 + CD8 T immunity decreased myeloid-derived suppressor cells (MDSCs) infiltration glioma. Collectively, restrained the progress possibly improving cell surveillance inhibiting MDSC microenvironment.
