Gladiolus is an important and economically valued ornamental plant grown worldwide. One of the major challenges in its micropropagation maintaining genetic stability during indirect regeneration long-term callus maintenance. The objective this study was to develop optimized shoot protocol for three commercial cultivars with consistent traits. The propagation of arabica coffee (Coffea L.) seedlings through indirect primary embryogenesis has significant potential to produce many healthy relatively quickly. However, there is a concern that plant growth regulators (PGRs) applied in the culture media could result somaclonal variations. To address this issue, appropriate concentration PGR needs be identified increase success somatic process while preventing This study aimed develop protocol for C. and evaluate variation plantlets derived from embryogenesis, considering genotype used. Three cultivars coffee, i.e., AS2K, S-795, Sigarar Utang, two types PGRs (2,4-D 2-iP) at different combination concentrations, were used as factorial experimental design. was 4.52 9.04 µM 2,4 D 4.93, 9.86, 14.79, 19.72, 24.65 2-iP. Fusarium wilt, caused by the fungus oxysporum f. sp. cubense (Foc), is one of most devastating diseases affecting banana cultivation worldwide. Although Foc tropical race 4 (TR4) has not yet been identified in Brazilian production areas, damage races 1 and subtropical main cause losses, especially cultivars Prata subgroup. Thus, induction somaclonal variation a promising strategy biotechnology to generate genetic variability develop resistant varieties. This study aimed induce Catarina cultivar (AAB genome) using successive subcultures Murashige Skoog (MS) medium enriched with plant regulator Thiadizuron (TDZ) at two concentrations: 2 mg/L. After evaluating symptoms, we selected 13 somaclones that were infected fungus. Histochemical histological analyses indicated possible defense mechanisms prevented colonization and/or infection Foc, such as intense phenolic compounds presence cellulose callose roots.
