Curcumin, a hydrophobic polyphenol extracted from the rhizome of Curcuma longa, is now considered candidate drug for treatment neurological diseases, including Parkinson’s Disease (PD), Alzheimer’s (AD), Huntington’s (HD), Multiple Sclerosis (MS), Amyotrophic Lateral (ALS), and prion disease, due to its potent anti-inflammatory, antioxidant potential, anticancerous, immunomodulatory, neuroprotective, antiproliferative, antibacterial activities. Traditionally, curcumin has been used medicinal dietary purposes in Asia, India, China. However, low water solubility, poor stability blood, high rate metabolism, limited bioavailability, little capability cross blood–brain barrier (BBB) have clinical application curcumin, despite important pharmacological activities this drug. Despite the diverse applications of γ radiation in radiotherapy, industrial processes, and sterilization, it causes hazardous effects on living organisms, such as cellular senescence, persistent cell cycle arrest, mitochondrial dysfunction. This study evaluated efficacy curcumin nanoparticles (CNPs) mitigating dysfunction senescence induced by rat brain tissues. Four groups male Wistar albino rats (n = 8 per group) were included: (Gr1) control group; (Gr2) CNPs group (healthy receiving oral administration at a dose 10 mg/kg/day, three times week for eight weeks); (Gr3) irradiated (rats exposed to single Gy head irradiation); (Gr4) + (irradiated treated with CNPs). The data obtained demonstrated that weeks attenuated oxidative stress γ-irradiated lowering brain's lipid peroxidation level [malondialdehyde (MDA)] enhancing antioxidant markers [superoxide dismutase (SOD), reduced glutathione (GSH), total capacity (TAC)] (P < 0.05). Background/Objectives: Epilepsy is a complex neurological disorder with strong link to oxidative stress, which contributes seizure susceptibility and neuronal damage. This study aims investigate the effects of curcumin (Cur), sodium valproate (VPA), mitocurcumin (MitoCur), mitochondria-targeted curcumin, on behavioral stress parameters in zebrafish model pentylenetetrazole (PTZ)-induced seizures. Methods: Adult were exposed two concentrations (0.25 0.5 µM for Cur MitoCur; 0.25 mM VPA). Behavioral assessments, including locomotion, spatial exploration, directional movement, conducted using EthoVision XT tracking software. Oxidative markers, superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GPx), total antioxidant status (TAS), analyzed brain homogenates. Results: analyses indicated dose-dependent effects, higher doses generally reducing activity. MitoCur at enhanced defenses reduced damage, while exhibited pro-oxidant shift. VPA improved TAS without significantly altering MDA levels. The topical administration of in situ hydrogels for ocular pathologies is a promising application strategy providing high effectiveness and patient compliance.
