Abstract Infectious disease is one of the major threats to humans and it second leading cause death worldwide. Edible mushrooms have many nutritional medicinal values human health. The properties edible mushroom extract in inhibiting pathogenic microorganisms had advantages over use chemically synthetic antimicrobial compounds due less unwanted side effects can combat microbial resistance. This study hypothesized that polarity affects extraction quality Hericium erinaceus fruiting bodies which was prepared subsequently its activity as an against six tested microorganisms, including MRSA, Streptococcus mutans , Enterobacter cloaca Salmonella typhimurium Candida lipolytica ; antiviral Hepatitis A virus (HAV) virus; antioxidant using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay; anti-inflammatory potential. There is a consensus that the antifungal repertoire for treatment of cryptococcal infections limited. Standard involves administration an drug derived from natural sources (i.e., amphotericin B) and two other drugs developed synthetically flucytosine fluconazole). Despite treatment, mortality rates associated with fungal cryptococcosis are high. Amphotericin B toxic, require intravenous administration, usually unavailable in low-income countries because their high cost. However, fluconazole cost-effective, widely available, harmless regard to its side effects. fungistatic agent has contributed considerably increase resistance frequent relapses patients meningitis. Therefore, there unquestionable need identify new alternatives or adjuvants conventional cryptococcosis. A potential should be able kill cryptococci "bypass" virulence mechanism yeast. Furthermore, it have fungicidal action, low toxicity, selectivity, easily penetrate central nervous system, available. In this review, we describe cryptococcosis, therapy, failures arising use traditionally considered reference standard. Additionally, present approaches used discovery counteract ranging screening products inclusion structural modifications optimize anticryptococcal activity, as well repositioning combined therapies.

Marine organisms, especially sponges, provide many sources of metabolites with various biological activities. Most sponges associate microbes such as fungi. To solve the problem sponge availability, it is necessary to isolate compounds from marine-derived fungi due their feasibility and advantages. This study, thus, highlights that most prominent genera produce active antibacterials antibiofilm were Aspergillus, Penicillium, Neosartorya, Trichoderma. The summarized data isolated related antibacterial activities until 2022 included anthraquinones, sterigmatocystin analogs, hydroxy pyrrolidine alkaloids, helvolic acid derivatives, lactones, prenylated phenylbutyrolactones, citrinin bisthiodiketopiperazine, cyclotetrapeptides, dihydrochromone dimers, amino lipopeptides, furan aspiron-derivatives, halogenated metabolites, alkaloids. Since biofilm mechanism very complex, some do not necessarily work antibiofilms. Nevertheless, can be concluded produced sponge-associated have potential developed new agents although still require further investigation actions compounds.

Marine sponges are noteworthy sources of bioactive secondary metabolites that demonstrate fundamental functions in defense mechanisms and ecological interactions. This study focused on the extraction characterization from two sponge species, Echinodictyum asperum Callyspongia siphonella, obtained Red Sea coast Saudi Arabia. Methanol generated EAE CSE extracts, which were subjected to metabolic profiling, illuminating 44 identified involving fatty acids, sterols, terpenoids, long-chain amides, alkaloids. The cytotoxic effects both assessed versus MDA-MB-231 MCF-7 breast cancer cell lines, utilizing sulforhodamine B (SRB) assay, with IC50 values about 2.7 μg/mL for 0.5 cells 8.7 45.3 cells. Flow cytometry analysis indicated extracts induced apoptosis necrosis across lines. Particularly, G2 phase arrest cells, while elevated S population As well, boosted autophagy especially Comparative analyses existing literature underscore therapeutic potential these sponge-derived medication. Molecular docking studies demonstrated robust binding affinities between selected key targets MCL-1, BCL-2, ERα. In conclusion, this research emphasizes significant features proposing their purposes treatment via distinct action.

Abstract Many lactic acid bacteria (LAB), known for their human health benefits, are derived from milk and utilized in biotherapeutic applications or producing valuable nutraceuticals. However, the specific role of milk-associated LAB biotherapeutics remains underexplored. To address this, eight product samples were randomly selected Egyptian market, diluted, then cultured anaerobically on MRS agar. Subsequently, 16 suspected isolates recovered underwent rapid preliminary identification. Among these isolates, Lactobacillus plantarum strain with accession number (OQ547261.1) was identified due to its strong antioxidant activity depending DPPH assay, L. displayed notable activities 71.8% 93.8% at concentrations 125–1000 µg/mL, respectively. While ascorbic showed lower 7.81, 3.9, 1.95 µg/mL which 45.1%, 34.2%, 27.2%, The anti-inflammatory efficacy evaluated based capability prevent hemolysis induced by hypotonic conditions. At a concentration 1000 could reduce 97.7%, nearly matching 99.5% inhibition rate achieved standard drug, indomethacin, an identical concentration. Moreover, exhibited high hemolytic 100 (14.3%), decreased 1.4% µg/mL. abundance phenolic acids flavonoids determined high-performance liquid chromatography (HPLC) . Real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) demonstrated that increased gene expression inflammatory marker TLR2 133%, cellular oxidation markers SOD1 SOD2 65% 74.2%, respectively, while suppressing CRP 33.3%. These results underscore plantarum’s exceptional activities. Furthermore, induces cancer cell death through necrotic nuclear DNA fragmentation. findings suggest is not only suitable nutraceutical production but also holds potential as probiotic strain. Future research should focus enhancing capacity this across various industries fostering innovation multiple fields.
