Mitochondrial dysfunction has been linked to psoriasis, and may be an important underlying factor contributing this disease. However, a precise methodology for assessing mitochondrial yet developed. One promising approach is measure NADH autofluorescence from affected skin areas. In study, we show that Flow Mediated Skin Fluorescence (FMSF) can used non-invasive assessment of in psoriasis. The fluorescence level at baseline the half-time ischemic growth (t1/2) derived FMSF traces NADH/NAD+ redox imbalance psoriatic lesions compared unaffected skin. These results are supported by analysis key parameters, Reactive Hyperemia Response (RHR) Hypoxia Sensitivity (HS). This method not only contributes understanding biochemical processes involved etiopathogenesis but also provides basis identifying new drug targets improving treatment process.

Flow Mediated Skin Fluorescence (FMSF) is a new non-invasive diagnostic method for assessing vascular circulation and/or metabolic regulation. The measures stimulation of the in response to post-occlusive reactive hyperemia (PORH). It analyzes dynamical changes NADH fluorescence emitted from skin tissue, providing information on mitochondrial status and intracellular oxygen delivery through circulatory system. Assessment state using FMSF technique based three parameters: flowmotion (FM) under normoxia conditions, hypoxia sensitivity (HS), hyperemic (HRmax). functioning mitochondria can be assessed by analyzing ischemic (IRmax), basal level fluorescence. There close relationship between Despite these interactions, regulatory function monitored separately as well simultaneously technique. Uniquely, this approach delivers both single measurement.

Adenine nucleotides play a critical role in maintaining essential functions of red blood cells (RBCs), including energy metabolism, redox status, shape fluctuations and RBC-dependent endothelial microvascular functions. Recently, it has been shown that infection with the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) might lead to morphological metabolic alterations erythrocytes both mild cases coronavirus disease (COVID-19). However, little is known about effects COVID-19 on nucleotide energetics RBCs nor potential contribution metabolism long COVID syndrome. This study aimed analyze levels adenine isolated from patients 12 weeks after SARS-CoV-2 who suffered symptoms relate them function parameters as well rate peripheral tissue oxygen supply. Although absolute quantities were rather slightly changed individuals, many related microcirculatory showed significant correlations RBC adenosine triphosphate (ATP) total (TAN) concentration. A particularly strong relationship was observed between ATP serum ratio arginine asymmetric dimethylarginine—an indicator function. Consistently, positive correlation also ATP/ADP diminished reactive hyperemic response patients, assessed by flow-mediated skin fluorescence (FMSF) technique, which reflected decreased vascular nitric oxide bioavailability. In addition, we have significantly impaired ischemic (IR max IR index), examined FMSF, revealed residual bioavailability epidermal keratinocytes brachial artery occlusion. These ratio, confirming key bioenergetics Taken together, outcomes this indicate dysregulation processes co-occurring dysfunction associated intracellular delivery, may partly explain COVID-specific such physical impairment fatigue.

Arterial blood pressure monitoring plays an important role in preventive medicine, allowing, selected cases, the identification of vascular dysfunction. In this review, we propose a new non-invasive approach to assessment circulatory system, based on its reaction hypoxia induced by post-occlusive reactive hyperemia (PORH). Three key parameters can be used for screening: Reactive Hyperemia Response (RHR), which represents overall macro- and microcirculation transient hypoxia; Hypoxia Sensitivity (HS), reflects hypoxia-induced activation myogenic oscillations microcirculation; Normoxia Oscillatory Index (NOI), characterizes microcirculatory under normoxia conditions. A method assessing these parameters, analogous simplicity arterial measurement, is provided Flow Mediated Skin Fluorescence (FMSF) technique. Reference values are proposed numerous test measurements.

Background: Erectile dysfunction (ED) most often has vascular etiology and usually is the earliest symptom of dysfunction. The aim this study was to evaluate with use Flow-Mediated Skin Fluorescence (FMSF) technique in men without ED. Methods: Included were 39 (median age 53) ED 40 41.5) Medical interview, physical examination, anthropometrical measurements performed for all participants. serum total testosterone, LH, SHBG determinations patients ED, Free Testosterone Index (FTI) calculated. FMSF used measure microcirculatory oscillations at baseline determine flowmotion (FM) vasomotion (VM) parameters. Normoxia Oscillatory (NOI) calculated, which represents contribution endothelial (ENDO) neurogenic (NEURO) relative detected low-frequency intervals (<0.15 Hz): NOI = (ENDO + NEURO)/(ENDO NEURO VM). Results: In found significantly lower FM VM parameters, but higher comparison correlated positively erectile function, orgasmic general sexual satisfaction whole group FTI group. thresholds 53.5 (AUC 0.7) 8.4 predictive values discriminating Conclusions: It shown that diagnostic may be helpful early diagnosis microcirculation due impaired caused by decreased testosterone activity.

Introduction . Diagnostics of blood circulation in skin microvessels and oxidative metabolism biological tissue allows predicting the presence microcirculatory disorders body, assessing their severity monitoring therapy. The use a functional test related to change position entire human body space is well-known method for studying functions autonomic nervous system diagnosing cardiovascular diseases. Purpose To study set parameters characterizing dynamics microcirculation finger during sequence tests with upper limb using fluorescence spectroscopy laser Doppler flowmetry. Materials Methods was conducted on group volunteers consisting 10 people aged 21–40 years. LAZMA PF device (Russia), implementing methods flowmetry spectroscopy, placed pad middle finger. Each volunteer’s were measured two arm positions: «at heart level» vertically raised «up» position. Blood flow oscillations continuously all stages: three times positions. Results A from state leads decrease average values indices by 12 perfusion units (p. u.) 7 relative (r. simultaneous increase NADH coenzyme 3 r. u., i.e. there unidirectional an oppositely directed values. Critical illness is often accompanied by a hemodynamic imbalance between macrocirculation and microcirculation, as well mitochondrial dysfunction. Microcirculatory disorders lead to abnormalities in the supply of oxygen tissue cells, while dysfunction leads abnormal energy metabolism impaired utilization, making these conditions important pathogenic factors critical illness. At same time, there close relationship microcirculation mitochondria. We introduce here concept “critical unit”, with two core components: which mainly comprises microvascular network endothelial especially glycocalyx; mitochondria, are involved but perform other non-negligible functions. This review also introduces several techniques devices that can be utilized for real-time synchronous monitoring thus unit monitoring. Finally, we put forward concepts strategies unit-guided treatment.

This review summarizes studies dedicated to the assessment of microvascular function based on microcirculatory oscillations monitored by FMSF technique. Two approaches are presented. The first approach uses oscillatory parameters measured under normoxia conditions, expressed as flowmotion (FM), vasomotion (VM), and index (NOI). These have been used for identification impaired associated with intense physical exercise, post-COVID syndrome, psychological stress, erectile dysfunction. second involves characterization response hypoxia measurement sensitivity (HS). HS parameter is characterize complications in diabetes, such diabetic kidney disease foot ulcers. Based research conducted authors this review, ranges characterizing diagnostic assessing technique has a wide range applications potential be integrated into widespread medical practice.
