The skin is constantly exposed to exogenous and endogenous sources of reactive oxygen species (ROS). An adequate balance between ROS levels antioxidant defenses necessary for the optimal cell tissue functions, especially skin, since it must face additional that do not affect other tissues, including UV radiation. Melanocytes are more oxidative stress than cells, also due melanin production process, which itself contributes generating ROS. There an increasing amount evidence may play a role in many diseases, melanoma, being primary cause or cofactor aggravates condition. Indeed, emerging as another major force involved all phases melanoma development, only arising malignancy but progression toward metastatic phenotype. Furthermore, seems chemoresistance thus has become target therapy. In this review, we discuss existing knowledge on examining defenses, giving particular consideration melanocytes. Therefore, focus significance analyzing possibility exploit induction therapeutic strategy improve effectiveness management melanoma.

Diabetic nephropathy is a common complication of type I and II diabetes, in which renal glomeruli are destroyed, resulting damage, proteinuria, hypertension. Apoptosis, autophagy, necroptosis 3 forms programmed cell death that have been implicated the pathogenesis diabetic nephropathy. Apoptosis podocytes leads to glomerular injury podocyte depletion, associated with proteinuria structural damage Additionally, epithelial cells proximal convoluted tubules also undergo apoptosis nephropathy, leading tubular atrophy, causes depletion subsequent formation atubular association loss function. On other hand, insufficiency autophagy has correlated For instance, decreased autophagic activity shown kidney, causing variations function disruption filtration barrier. Furthermore, attenuated demonstrated buildup impaired molecules organelles, normally broken down by proteinuria. Moreover, might key role decline Thus, this article aims review mechanisms effects including roles apoptosis, necroptosis.

The role of the tumor microenvironment in growth and therapy has recently attracted more attention research drug development. ability to trigger maintenance, progression, resistance is main cause for treatment failure relapse. Accumulated evidence indicates that maintenance progression cells determined by components microenvironment, which include stromal (endothelial cells, fibroblasts, mesenchymal stem immune cells), extracellular matrix (ECM), soluble molecules (chemokines, cytokines, factors, vesicles). As a solid tumor, melanoma not only mass monolithic but it also contains supporting stroma, ECM, molecules. Melanoma are continuously interaction with microenvironment. In present review, we focus on modulation as well impact therapeutic target melanoma.

Abstract Multi-modal combination therapy is regarded as a promising approach to cancer treatment. Combining chemotherapy and phototherapy an essential multi-modal endeavor. Ivermectin (IVM) potent antiparasitic agent identified having potential antitumor properties. However, the fact that it induces protective autophagy while killing tumor cells poses challenge its further application. IR780 iodide (IR780) near-infrared (NIR) dye with outstanding photothermal (PTT) photodynamic (PDT) effects. hydrophobicity, instability, low uptake of limit clinical applications. Here, we have structurally modified hydroxychloroquine, inhibitor, synthesize novel compound H780. H780 IVM can form H780-IVM nanoparticles (H-I NPs) via self-assembly. Using hyaluronic acid (HA) modify H-I NPs, nano-delivery system HA/H780-IVM (HA/H-I was synthesized for chemotherapy-phototherapy colorectal (CRC). Under NIR laser irradiation, HA/H-I NPs effectively overcame limitations exhibited cytotoxicity. In vitro in vivo experiment results showed excellent anti-CRC Therefore, our study provides strategy CRC treatment could enhance chemo-phototherapy by modulating autophagy.

Rheumatoid arthritis (RA) is a chronic, progressive, systemic autoimmune disease characterised by synovial inflammation, pannus formation and subsequent destruction of articular cartilage bone. Programmed cell death (PCD), encompassing apoptosis, autophagy, pyroptosis, necroptosis, ferroptosis, plays pivotal role in the pathogenesis RA. An imbalance PCD causes variety immune cells to release large amounts inflammatory factors mediators that exacerbate not only chronic but also bone joint damage. The purpose this article review relevant studies between RA, with aim providing further insights considerations for deeper understanding RA guide clinical management.

Abstract Breast cancer has become the most prevalent cancer, globally. Adriamycin is a first-line chemotherapeutic agent, however, cells acquire resistance to it, which one of common causes treatment failure. ROS and NRF2 are essential oxidative stress factors that play key role in process associated with cancer. Our goal create novel therapeutic drugs or chemical sensitizers will improve chemotherapy sensitivity. The optimal concentration duration for MCF-7 MCF-7/ADR ADR CYT were determined using CCK-8 assay. We found + inhibited activity breast as well causing apoptosis blocking cell cycle G0/G1 phase. induces through generation P62/NRF2/HO-1 signaling pathway. In cancer-bearing nude mice, effectively suppressed tumor development vivo. Overall, our findings showed combination potent anti-breast both vivo vitro, suggesting main drug used chemosensitivity.

This research aims to examine the transdermal release of water-soluble indium and zinc metallo phthalocyanine (InPc ZnPc) compounds from poly(vinyl alcohol) (PVA) membrane cytotoxicity effect these Pcs on normal mouse fibroblasts (L929 fibroblast) human melanoma (SK-MEL-30) cells. For this purpose, effects temperature, pH, drug concentration thickness were investigated in order obtain optimum profile by preparing PVA membranes with different thicknesses crosslinked heat treatment. Optimum was found be 85.36% using 6 μm thick at 37 ± 0.5 °C, when upper cell pH 1.2 lower 5.5, for 3 mg/mL InPc concentration. Under same conditions, value ZnPc 69.78%. In addition, vitro studies performed L929 SK-MEL-30 under optimized conditions. It that no significant cytotoxic observed cells dark. Photodynamic tests also carried out ZnPc. The results show viability decreases concentrations 10 μg/mL above. while IC50 determined as 4.058 μg/mL, 11.574
