Spinal Cord Injury (SCI) is a common neurological disorder with devastating psychical and psychosocial sequelae. The majority of patients after SCI suffer from permanent disability caused by motor dysfunction, impaired sensation, neuropathic pain, spasticity as well urinary complications, small number experience complete recovery. Current standard treatment modalities the aim to prevent secondary injury provide limited recovery lost functions. Stem Cell Therapy (SCT) represents an emerging approach using differentiation, paracrine, self-renewal capabilities stem cells regenerate injured spinal cord. To date, multipotent including mesenchymal (MSCs), neural (NSCs), hematopoietic (HSCs) represent most investigated types for in preclinical clinical studies. microenvironment has significant impact on survival, proliferation, differentiation transplanted cells. Therefore, deep understanding pathophysiology molecular mechanisms through which act may help improve efficacy SCT find new therapeutic approaches such stem-cell-derived exosomes, gene-modified cells, scaffolds, nanomaterials. Abstract Chronic pain constitutes one of the most common chronic complaints that people experience. According to International Association for Study Pain, is defined as persists or recurs longer than 3 months. has a significant impact on individuals’ well-being and psychosocial health economy healthcare systems well. Despite availability numerous therapeutic modalities, treatment can be challenging. Only about 30% individuals with non-cancer achieve improvement from standard pharmacological treatment. Therefore, approaches were proposed potential including non-opioid agents, nerve blocks, acupuncture, cannabidiol, stem cells, exosomes, neurostimulation techniques. Although some methods such spinal cord stimulation successfully introduced into clinical practice therapy pain, current evidence brain efficacy in remains unclear. Spinal cord injury (SCI) disrupts nerve pathways and affects sensory, motor, autonomic function. There is currently no effective treatment for SCI. SCI occurs within three temporal periods: acute, subacute, chronic. In each period there are different alterations in the cells, inflammatory factors, signaling spinal cord. Many biomaterials have been investigated of SCI, including hydrogels fiber scaffolds, some progress has made using multiple materials. Although many studies have investigated the role of cytokines in bone metastases, our knowledge their function spine metastasis is limited. Patients suffering from painful spinal bone metastases (PSBMs) often undergo palliative radiation therapy (RT), with an efficacy of approximately two thirds patients. In this exploratory investigation, we assessed the effectiveness machine learning (ML) models trained on radiomics, semantic and clinical features to estimate complete pain response. Gross tumour volumes (GTV) target (CTV) 261 PSBMs were segmented planning computed tomography (CT) scans. Radiomics, collected for all Random forest (RFC) support vector (SVM) classifiers compared using repeated nested cross-validation. The best radiomics classifier was CTV area under receiver-operator curve (AUROC) 0.62 ± 0.01 (RFC; 95% confidence interval). model achieved a comparable AUROC 0.63 (RFC), significantly below (SVM, AUROC: 0.80 0.01); slightly lower than instability neoplastic score (SINS; LR, 0.65 0.01). A combined did not improve performance (AUROC: 0,74 0,01). We could demonstrate that analyses CTs allowed limited prediction response RT. ML predictions based established parameters results.

Abstract Spinal tumours are uncommon lesions, but they may cause significant morbidity and mortality.
