Obesity represents a serious medical condition and has nowadays reached pandemic proportions. Comorbidities associated with obesity are severe, affect almost all organ systems. Adipose tissue several important functions: to provide energy storage, maintain body temperature through the process of thermogenesis, balance in variety metabolic patterns. is an endocrine numerous biochemical roles, obese individuals adipose becomes either hypertrophic or hyperplastic, impaired function. The immune system activity regulation extremely related processes. A loss on this axis causes disorders obesity, basis these insulin resistance, which hallmark obesity. Hormones that exclusively produced adipocytes studied various biochemistry processes leptin, adiponectin resistin. Successful treatment involves lifestyle diet modification, pharmacotherapy, mainly glucagon-like peptide-1 agonists, bariatric surgery, remains most effective method for selected population severely individuals. Positive effects surgery reflected not only restriction food intake, but hormonal changes, especially those gut hormones responsible hunger metabolism.

We sought to advance our understanding of skeletal muscle lipid metabolism and dynamics in cancer cachexia. Cachexia increased the number size intramyocellular droplets (LDs). Furthermore, decreases LD-mitochondrial touch, contact length, relative along with LD shape complexity circularity roundness. Dysregulation LD-associated proteins was also documented. Collectively, we show that myosteatosis, altered morphology, decreased interactions occur

Lipid droplets (LDs), highly dynamic cellular organelles specialized in lipid storage and maintenance of homeostasis, contain several proteins on their surface, among which the perilipin (Plin) family stands out as most abundant group LD-binding proteins. They play a pivotal role influencing behavior functionality LDs, regulating lipase activity, preserving balance between synthesis degradation, is crucial development obesity abnormal accumulation fat non-adipose tissues, causing negative adverse biological effects, such insulin resistance, mitochondrial dysfunction, inflammation. The expression levels Plins are often associated with various diseases, hepatic steatosis atherosclerotic plaque formation. Thus, it becomes interest to investigate Plin roles by using appropriate "omics" approaches that may provide additional insight into mechanisms through these contribute tissue homeostasis. This review intended give an overview significant omics studies focused characterization identification potential targets involved progression cardiovascular cardiometabolic complications, well interactors could be useful for more efficient therapeutic preventive patients.

Abstract Glioma is a common and destructive brain tumor, which highly heterogeneous with poor prognosis. Developing diagnostic prognostic markers to identify treat glioma early would significantly improve the therapeutic outcomes. Here, we conducted RNA next-generation sequencing 33 samples 15 normal samples. We found Perilipin 1 (PLIN1) downregulated in correlated poorer outcome. Subsequent experiments revealed that up regulation of PLIN1 led repressed cell growth invasion glioma. Moreover, overexpression increased lipid accumulation cells, increasing expression biosynthesis related genes decreasing lipolysis genes. Mechanically, PI3K/AKT axis could regulate levels glioma, inhibition activity increase In conclusion, dysregulation downregulation following tumor proliferation, metabolism reprogramming

Cancer-associated fibroblasts are an essential part of the tumor immunoenvironment, playing key roles in malignancy progression and treatment response. This study was to characterize cancer-associated fibroblasts-related genes (CAFs) colorectal cancer (CRC) establish signature associated with CAF for prognosis prediction. We downloaded single-cell RNA sequencing (scRNA-seq) data from GEO database bulk RNA-seq TCGA identify differentially expressed related fibroblasts. In set, DEGs were identified samples, WGCNA method utilized module genes. By comparing fibroblast-related DEGs, we took overlapped cohorts as crucial CAFs. Moreover, prognostic CAFs using univariate analysis. A risk model established LASSO algorithm then validated external datasets. Ultimately, expression CRC confirmed qRT-PCR. large cohort CAFs, eight demonstrating significance. These primarily seven pathways, including peroxisome function, B cell receptor signal, adhesion molecule. The exhibited high accuracy predicting prognosis, through validation independent cohorts. Additionally, showed significant correlations immune-related scores, purity, estimate, stromal scores. qRT-PCR that level RAB36 significantly downregulated HCT116 HT29 lines compared NCM460 cells. Conversely, CD177, PBX4 CCDC78 upregulated lines, ACSL6 KCNJ14 only cells (P < 0.05). trends CD177 consistent our predicted results. accurately immune infiltration, estimates. Over the past 10 years, biological role of lipid droplets (LDs) has gained significant attention in context both physiological and pathological conditions. Considerable progress been made elucidating key aspects these organelles, yet much remains to be accomplished fully comprehend myriad functions they serve progression hepatic tumors. Our current perception is that LDs are complex active structures managed by a distinct set cellular processes. This understanding represents paradigm shift from earlier perspectives. In this review, we aim recapitulate function within liver, highlighting their pivotal pathogenesis metabolic dysfunction-associated steatotic liver disease (MASLD) (Hsu Loomba, 2024) contribution towards more advanced stages up hepatocellular carcinoma (HC) (Farese Walther, 2009). We aware molecular complexity changes occurring neoplastic evolution liver. attempt, however, summarize most important recent roles across healthy all states, hepatocarcinoma.
