Cannabinoids: cannabidiol (CBD), cannabidiolic acid (CBDA), and cannabichromene (CBC) are lipophilic compounds with limited water solubility, resulting in challenges related to their bioavailability therapeutic efficacy upon oral administration. To overcome these limitations, we developed co-dispersion cannabinoid delivery systems the biopolymer polyvinyl caprolactam-polyvinyl acetate-polyethylene glycol (Soluplus) magnesium aluminometasilicate (Neusilin US2) improve solubility permeability. Recognizing potential benefits arising from entourage effect, decided work an extract instead of isolated cannabinoids. Cannabis sativa inflorescences (Henola variety) a confirming neuroprotective activity were subjected dynamic supercritical CO2 (scCO2) extraction next they combined carriers (1:1 mass ratio) prepare (HiE). In vitro dissolution studies conducted evaluate CBD, CBDA, CBC various media (pH 1.2, 6.8, fasted, fed state simulated intestinal fluid). The HiE-Soluplus consistently demonstrated highest rate Additionally, exhibited permeability coefficients for cannabinoids gastrointestinal tract conditions than it was during using model PAMPA GIT. All three promising blood-brain barrier (BBB) (Papp higher 4.0 × 10−6 cm/s), suggesting effectively cross into central nervous system. improved system hold promise enhancement bioavailability.

The endoplasmic reticulum (ER) is the organelle mainly involved in maintaining cellular homeostasis and driving correct protein folding. ER-dependent defects or dysfunctions are associated with genesis/progression of several pathological conditions, including cancer, inflammation, neurodegenerative disorders, that directly indirectly correlated to a wide set events collectively named under term "ER stress". Despite recent increase interest concerning ER activity, further research studies needed highlight all mechanisms responsible for failure. Introduction: This study aimed to analyze the effects of cannabis oil (cannabidiol:tetrahydrocannabinol [CBD:THC], 2:1 ratio) on mechanisms involved in hepatic steatosis and oxidative stress an experimental model metabolic syndrome (MS) induced by a sucrose-rich diet (SRD). We hypothesized that noninvasive oral administration improves through lower activity lipogenic enzymes increase carnitine palmitoyltransferase-1 (CPT-1) enzyme mitochondrial oxidation fatty acids. Furthermore, ameliorates liver regulation main regulatory factors involved, nuclear factor erythroid 2 (NrF2) factor-kB (NF-κB) p65. For testing this hypothesize, relevant MS was feeding rats with SRD for 3 weeks. Methods: Male Wistar were fed following diets weeks: reference diet: standard commercial laboratory diet, SRD, + oil: 1 mg/kg body weight daily. The full-spectrum presents total cannabinoid CBD:THC ratio. Serum glucose, triglyceride, cholesterol, HDL-cholesterol, LDL-cholesterol, uric acid, alanine aminotransferase, aspartate alkaline phosphatase (AP), N-arachidonoylethanolamine or anandamide 2-arachidonoylglycerol endocannabinoids levels, thiobarbituric acid reactive substance (TBARS) non-enzymatic antioxidant capacity (ferric ion-reducing power [FRAP]) evaluated. In tissue: histology, nonalcoholic disease score (NAS), triglycerides cholesterol content, activities (fatty synthase, acetyl-CoA carboxylase, malic enzyme, glucose-6-phosphate dehydrogenase), related (CPT-1), oxygen species, TBARS, FRAP, glutathione, catalase, glutathione peroxidase, reductase activities. 4-hydroxynonenal, NrF2, NF-κB p65 levels analyzed immunohistochemistry. Results: results showed SRD-fed developed dyslipidemia, damage, (increase key novo synthesis decrease oxidation), lipid peroxidation, stress. Hepatic NrF2 expression significantly decreased increased. Cannabis improved steatosis, peroxidation (improving metabolism), tissue, increased, reduced. Conclusion: present revealed new aspects damage dyslipidemic insulin-resistant rats. demonstrated properties molecular (CBD:THC, lipotoxicity MS.
