Successful engineering of functional salivary glands necessitates the creation cell‐instructive environments for ex vivo expansion and lineage specification primary human gland stem cells (hS/PCs). Herein, basement membrane mimetic hydrogels are prepared using hyaluronic acid, cell adhesive peptides, hyperbranched polyglycerol (HPG), with or without sulfate groups, to produce “hyperGel+” “hyperGel”, respectively. Differential scanning fluorescence experiments confirm ability sulfated HPG precursor stabilize fibroblast growth factor 10. The nanoporous, cytocompatible, cell‐permissive, enabling development multicellular hS/PC spheroids in 14 days. incorporation species hydrogel enhances proliferation. Culture hS/PCs hyperGel+ presence a Rho kinase inhibitor Y‐27632 (Y‐27) leads central lumen, increases expression acinar marker aquaporin‐3 at transcript level ( AQP3 ), decreases ductal keratin 7 both KRT7 ) protein levels (K7). Reduced transforming beta (TGF‐β) targets SMAD2/3 is also observed Y27‐treated cultures, suggesting attenuation TGF‐β signaling. Thus, cooperates Rho‐associated promote lumened enhanced markers.

Sjögren's syndrome (SS) is a chronic autoimmune disorder that primarily affects the exocrine glands. Due to intricate nature of disease progression, exact mechanisms underlying SS are not completely understood. Recent research has highlighted complex interplay between immune dysregulation and metabolic abnormalities in inflammatory diseases. Notably, lipid metabolism emerged as crucial factor modulation function progression diseases, including SS. This review explores prevalence dyslipidemia SS, emphasizing its role onset, prognosis disease. We specifically described impact altered glands association with disease-specific features, inflammation glandular dysfunction. Additionally, we discussed potential clinical implications regulation, polyunsaturated fatty acids (PUFAs) their deficits pathogenesis. By identifying promising therapeutic target, this highlights need for further into lipid-based interventions management

Sjogren's syndrome (SS) is a chronic autoimmune disorder that affects exocrine glands, particularly lacrimal leading to dry eye disease (DED). DED common ocular surface millions of people worldwide, causing discomfort, visual impairment, and even blindness in severe cases. However, there no definitive cure for DED, existing treatments primarily relieve symptoms. Consequently, an urgent need innovative therapeutic strategies based on the pathophysiology DED. Mesenchymal stem cells (MSCs) have emerged as promising tool various disorders, including SS-related (SS-DED). A intriguing facet MSCs their ability produce extracellular vesicles (EVs), which contain bioactive components such proteins, lipids, nucleic acids. These molecules play key role facilitating communication between modulating wide range biological processes. Importantly, MSC-derived EVs (MSC-EVs) properties similar those parent cells, immunomodulatory, anti-inflammatory, regenerative properties. In addition, MSC-EVs offer several notable advantages over intact MSCs, lower immunogenicity, reduced risk tumorigenicity, greater convenience terms storage transport. this review, we elucidate underlying mechanisms SS-DED discuss relevant targets treating SS-DED. comprehensively review broader landscape EV application corneal diseases. This focuses efficacy SS-DED, field study holds considerable appeal due its multifaceted regulation immune responses functions.

Sjögren’s syndrome is a multisystemic autoimmune disease that mainly affects the exocrine glands, causing dryness of eyes and mouth as principal symptoms. Non-coding RNAs (ncRNAs), once regarded genomic “junk”, are now appreciated important molecular regulators gene expression, not least in other diseases. Here we review research into causative roles microRNAs (miRNAs), long non-coding (lncRNAs), circular (circRNAs) on immunological responses, inflammation, salivary gland epithelial cell function patients. These ncRNAs represent promising new therapeutic targets for treating possibly biomarkers early diagnosis.

To assess the relationship between endothelial dysfunction and serum cytokines, anti-SSA anti-SSB antibodies, beta-2 microglobulin levels, focus score EULAR Sjögren’s Syndrome Disease Activity Index (ESSDAI) in primary syndrome (pSS) patients. The study included 90 patients with pSS 45 healthy subjects, matched for age gender, as controls. Serum microglobulin, total cholesterol, HDL-cholesterol, triglycerides, TNF-α, IL-6 were analyzed both groups. Patients also tested antinuclear anti-SAA (anti-Sjögren’s syndrome-related antigen A) (anti-Sjögren related B) (the histopathologic one, based on minor salivary gland biopsy). Endothelial was assessed by means of flow-mediated dilation (FMD) brachial artery. Data are presented mean ± standard deviation. Statistical analysis performed using t-test Pearson’s correlation. Differences considered to be statistically significant if value p < 0.05. identified (p 0.00001). levels cytokines (TNF-α, respective IL-6) increased compared controls (expressed FMD%) correlated score, ESSDAI, IL-6, statistical significance. is present associated a high activity well concentrations pro-inflammatory beta 2-microglobulin.
