A similar trend is emerging with root repair materials; however, there a significant gap in the literature about cementogenic ability bioceramic on periodontal ligament cells. The aim present study was to investigate effect (Biodentine and Bio-C Repair) cementogenesis potential stem cells (PDLSCs). Methods: PDLSCs were isolated using enzymatic digestion approach from sound extracted teeth. Material extracts prepared rubber discs immersed fresh growth medium 24 h at 37 °C. Reverse transcription–quantitative polymerase chain reaction (RT-qPCR) used detect mRNA expression levels markers cementum protein 1 (CEMP1), Cementum attachment (CAP), pathway transforming factor β1(TGF-β1), bone morphogenic 2 (BMP2), inflammatory marker IL-6. Results: Both showed significantly higher gene expressions when compared control groups. Repair Biodentine, except TGF-β1 expression, where both exhibited results. Conclusions: demonstrated Biodentine under tested conditions. Further vivo studies deemed necessary translate findings this into clinical practice.

Periodontal disease is a chronic inflammatory condition that destroys the tooth-supporting structures due to host's immune response microbial biofilms. Traditional periodontal treatments, such as scaling and root planing, pharmacological interventions, surgical procedures, have significant limitations, including difficulty accessing deep pockets, biofilm recolonization, development of antibiotic resistance. In light these challenges, natural bioactive compounds derived from plants, herbs, other sources offer promising alternative their anti-inflammatory, antioxidant, antimicrobial, tissue-regenerative properties. This review focuses on molecular mechanisms through which compounds, curcumin, resveratrol, epigallocatechin gallate (EGCG), baicalin, carvacrol, berberine, essential oils, Gum Arabic, exert therapeutic effects in disease. Bioactive inhibit critical pathways like NF-κB, JAK/STAT, MAPK while activating protective Nrf2/ARE, reducing cytokine production oxidative stress. They also activity matrix metalloproteinases (MMPs), preventing tissue degradation promoting healing. addition, demonstrated potential disrupt bacterial biofilms by interfering with quorum sensing, targeting cell membranes, enhancing efficacy.Bioactive modulate system shifting balance pro-inflammatory anti-inflammatory responses efferocytosis, helps resolve inflammation supports regeneration. Periodontitis, a chronic inflammatory disease triggered by dental plaque, often presents challenges in management, particularly severe cases where mechanical debridement alone may be insufficient. As result, adjunctive therapies, localized drug delivery systems with both antimicrobial and anti-inflammatory properties, are essential to enhance the efficacy of periodontitis management. In this study, we developed multifunctional hydrogel incorporating salicylic acid-choline deep eutectic solvent (DES) into chitosan/β-glycerol phosphate sodium (CS/GP) matrix for treatment periodontitis. The DES-CS/GP demonstrated favorable physicochemical including gelation injectability, making it highly suitable application oral cavity. effectively inhibited growth key periodontal pathogens, Porphyromonas gingivalis Fusobacterium nucleatum, significantly downregulated expression pro-inflammatory cytokines TNF-α IL-1β vitro. Cytocompatibility assessments showed over 80% cell viability human gingival fibroblasts, epithelial cells, keratinocytes 5 days treated DES-CS/GP, fluorescence microscopy confirming robust cytoskeletal integrity. Furthermore, enhanced permeability through tissues rat model periodontitis, mitigated bone loss, reduced bacterial loads P. g, suppressed tissues. These findings underscore hydrogel's potential as safe effective therapy offering combination antimicrobial, anti-inflammatory, tissue-permeating properties high biosafety ease application.

Matrix metalloproteinases (MMPs) are key enzymes involved in extracellular matrix (ECM) remodeling, regulating a wide range of cellular and immune processes both homeostatic pathological conditions. Host–microbiota interactions play critical role maintaining ECM balance; however, during dysbiosis, this regulation is disrupted, leading to compromised barrier integrity, pathogen translocation into circulation, the development systemic diseases cancer. This review highlights bidirectional relationship between MMP expression/activity microbiota emphasizing tissue-specific alterations activity that contribute disease progression. In addition, it integrates interdisciplinary evidence illustrate MMP-dependent mechanisms underlying various pathologies associated with oral gut microbiome including long-range effects through gut–skin gut–brain axes. Thus, introduces emerging field MatrixBiome, which explores complex ECM, microbiota, host tissues. Finally, also outlines therapeutic strategies modulate levels, either indirectly microbiome-targeted approaches (e.g., prebiotics, probiotics, postbiotics) or directly using inhibitors, offering promising avenues for future clinical interventions.

Background/Objectives: Previous research indicates that non-surgical periodontal therapy (NSPT) improves glycemic control in individuals with prediabetes and diabetes who have periodontitis. Few studies demonstrated its effects on mouthrinse active-matrix metalloproteinase-8 (aMMP-8) levels as it relates to control. We assessed the treatment response of stage II–IV periodontitis patients prediabetes, diabetes, normoglycemia, regarding glycated hemoglobin (HbA1c) aMMP-8 using point-of-care kits (PoC). Materials Methods: Eighty-eight adults (11 normoglycemic, 32 prediabetic, 45 type 2 diabetes), aged 25–78, were included. Full-mouth clinical examinations used evaluate their parameters. Periodontitis is a chronic inflammation caused by periodontopathogenic bacteria in the dental biofilm, and also involves inflammatory-immune response of host. Polymorphonuclear neutrophils (PMNs) play essential roles bacterial clearance multiple mechanisms, including formation neutrophil extracellular traps (NETs) that retain destroy pathogens. During PD progression, interaction between PMNs, NETs, leads to an exaggerated immune prolonged inflammatory state. As lesion matures, PMNs accumulate periodontal tissues die via NETosis, ultimately resulting tissue injury. A better understanding role associated molecules, pathogenic pathways NET periodontitis, could provide markers NETosis as reliable diagnostic prognostic tools. Moreover, assessment biomarker levels biofluids, particularly saliva or gingival crevicular fluid, be useful for monitoring periodontitis progression treatment efficacy. Preventing excessive accumulation tissues, both controlling NETs' their appropriate removal, key further development more efficient therapeutic approaches. In therapy, local drug delivery (LDD) systems are targeted, enhancing bioavailability active pharmacological agents pocket surrounding time ensure optimal outcome.
