Abstract Aims Anxiety often coexists with migraine, and both conditions share a commonality in oxidative/nitrosative stress mitochondrial dysfunction contributing to their pathogenesis. β‐Sitosterol, plant sterol, has shown promise mitigating stress, enhancing function, exerting neuroprotective effects. In this study, we investigated the impact of β‐sitosterol on migraine‐associated anxiety whether effect was associated alleviation improvement function. Methods Nitroglycerin used induce migraine adult male Wistar rats. β‐Sitosterol treatment consisted daily intraperitoneal injections (10 mg/kg) for 10 days following induction. levels were evaluated using open‐field test (OFT) hole‐board (HBT). Frontal cortex samples analyzed malondialdehyde (MDA), glutathione (GSH), reactive oxygen/nitrogen species, nitric oxide (NO) (markers stress), ATP (indicator function). Results Migraine induction led impaired performance OFT HBT. Concurrently, it elevated MDA, NO while diminishing GSH frontal cortex, signifying heightened stress. Moreover, decreased, indicating dysfunction. Treatment significantly restored behavioral assays normalized GSH, NO, ATP. Conclusion exerted anxiolytic effects which can be attributed its ability ameliorate enhance

PurposeThe pathophysiological mechanisms underlying migraine remain elusive, with oxidative stress hypothesized as a potential etiological factor. The Oxidative Balance Score (OBS) is comprehensive tool for assessing the impact of diet and lifestyle on stress, thereby gauging an individual's overall antioxidant capacity. In this cross-sectional study, we explored correlation between OBS prevalence among cohort US adults.MethodsWe analyzed data from 6195 participants aged 20 years above, drawn National Health Nutrition Examination Survey (NHANES) conducted 1999 2004. We employed multiple logistic regression, coupled sensitivity analyses, to investigate relationship migraine. Subsequent subgroup analyses interaction tests were performed assess consistency association across population.ResultsMultiple regression revealed inverse likelihood experiencing migraines. Specifically, individuals in highest quartile exhibited significantly reduced risk compared those lowest (OR = 0.98, 95% Confidence Interval (CI): 0.97–0.99, P 0.0001). Furthermore, restricted cubic spline curves indicated non-linear dietary incidence (non-linear 0.0258).DiscussionOur findings suggest that adherence antioxidant-rich may be effective strategy mitigating migraine, potentially by influencing balance.

Abstract Background Migraine stands as a prevalent primary headache disorder, with prior research highlighting the significant involvement of oxidative stress and inflammatory pathways in its pathogenesis chronicity. Existing evidence indicates capacity Dl-3-n-butylphthalide (NBP) to mitigate inflammation, thereby conferring neuroprotective benefits many central nervous system diseases. However, specific therapeutic implications NBP context migraine remain be elucidated. Methods We established C57BL/6 mouse model chronic (CM) using recurrent intraperitoneal injections nitroglycerin (NTG, 10 mg/kg), prophylactic treatment was simulated by administering (30 mg/kg, 60 120 mg/kg) gavage each NTG injection. Mechanical threshold assessed von Frey fibers, photophobia anxious behaviours were light/dark box elevated plus maze. Expression c-Fos, calcitonin gene-related peptide (CGRP), Nucleus factor erythroid 2-related 2 (Nrf2) related pathway proteins spinal trigeminal nucleus caudalis (SP5C) detected Western blotting (WB) or immunofluorescence (IF). The expression IL-1β, IL-6, TNF-α, Superoxide dismutase (SOD) malondialdehyde (MDA) SP5C CGRP plasma ELISA. A reactive oxygen species (ROS) probe used detect ROS SP5C. Results At end modelling period, mice showed significantly reduced mechanical nociceptive thresholds, well photophobic behaviours. Pretreatment attenuated sensitization, photophobia, anxiety mice, levels c-Fos activated Nrf2 downstream HO-1 NQO-1. By measuring associated cytokines, we also found that inflammation. Most importantly, effect after administration ML385 inhibit Nrf2. Conclusions Our data suggest may alleviate activating reduce inflammation models, confirming it potential drug for migraine. Cortical spreading depression (CSD) is a slow wave of cortical depolarization closely associated with migraines an aura. Previously, it was thought that CSD mainly driven by neurons, characteristic changes in neuronal swelling and increased extracellular potassium (K+) glutamate. However, the role astrocytes, member neurovascular unit, migraine has recently received increasing attention. In early stages CSD, astrocytes provide neurons energy support clear K+ glutamate from synaptic gaps. late release large amounts lactic acid to exacerbate hypoxia when demand exceeds astrocytes' compensatory capacity. Astrocyte endfoot are not similarly altered. It primarily due influx abnormally active calcium (Ca2+) signaling. Aquaporin 4 (AQP-4) only mediates little as aquaporin. Astrocytes causes perivascular space closure, slowing glymphatic system flow exacerbating neuroinflammation, leading persistent CSD. double-edged swords may be potential targets for interventions.
