Introduction: Aged-related brain damage and gut microbiome disruption are common. Research affirms that modulating the microbiota-gut-brain axis can help reduce age-related damage. Methods: Ginseng, esteemed in traditional Chinese medicine, is recognized for its anti-aging capabilities. However, previous Ginseng studies have largely focused on diseased animal models. To this end, efforts were hereby made to explore potential neuroprotective effects of fecal microbiota transplantation (FMT) from Ginseng-supplemented aged mice those pre-treated with antibiotics. Results: As a result, FMT specific modifications natural aging improved weight gain, extended telomere length, anti-oxidative stress tissue, regulated serum levels cytokine, balanced proportion Treg cells. Besides, increased abundance beneficial bacteria Lachnospiraceae , Dubosiella Bacteroides etc. decreased pathogenic Helicobacter Lachnoclostridium samples mice. This revealed remarkably reshaped microbiome. Additionally, FMT-treated showed metabolites Ursolic acid, β-carotene, S-Adenosylmethionine, Spermidine, Guanosine, Celecoxib, Linoleic etc., which significantly positively correlated critical above. these identified mainly enriched pathways Amino acid metabolism, Lipid Nucleotide Furthermore, downregulated p53/p21/Rb signaling upregulated p16/p14, ATM/synapsin I/synaptophysin/PSD95, CREB/ERK/AKT following aging. This study explores the potential of ginseng-derived peptides (GPs) as multifunctional bioactive agents for skincare. Unlike previous research into ginseng saponins and polysaccharides, we identified that extracts containing water-soluble small molecules polypeptides exhibit potent antioxidant, anti-inflammatory, anti-aging properties. In vitro assays revealed peptide extract (GPE) reduced reactive oxygen species (ROS) inflammatory cytokines (IL-6, TNF-α, IL-1β) in RAW264.7 macrophages while enhancing collagen synthesis human skin fibroblasts (HSFs). Validation using 3D epidermal dermal models further confirmed GPE’s ability to mitigate UV-induced damage, restore barrier proteins (filaggrin, loricrin), increase content. addition, screened 19 candidate from machine learning prioritized their interaction with aging inflammation-related targets. Three (QEGIYPNNDLYRPK, VDCPTDDATDDYRLK, ADEVVHHPLDKSSEVE) demonstrated significant collagen-promoting, anti-inflammatory effects cellular models. The pathogenesis of age-related macular degeneration (AMD), a degenerative retinopathy, remains unclear. Administration anti-vascular endothelial growth factor agents, antioxidants, fundus lasers, photodynamic therapy, and transpupillary warming has proven effective in alleviating symptoms; however, these interventions cannot prevent or reverse AMD. Increasing evidence suggests that AMD risk is linked to changes the composition, abundance, diversity gut microbiota (GM). Activation multiple signaling pathways by GM metabolites, including lipopolysaccharides, oxysterols, short-chain fatty acids (SCFAs), bile (BAs), influences retinal physiology. Traditional Chinese medicine (TCM), known for its multi-component multi-target advantages, can help treat altering composition regulating levels certain substances, such as reducing increasing SCFA BA contents. Mountain cultivated ginseng (MCG) is planted in mountain forests to simulate traditional wild ginseng; therefore, it has a greater pharmacological effect than (CG) the garden; however, insufficient evidence confirms this theory. In light of health-promoting and life-extending properties ginseng, we analyzed efficacy MCG CG. Initial observations revealed that phytosterols content was higher CG, with positive correlation duration growth. The distinction between CG predominately determined by stigmasterol using High-Performance Liquid Chromatography (HPLC). lifespan Drosophila melanogaster (fruit flies) aged naturally prolonged stigmasterols. Further, they healthy ageing as measured progeny numbers, length sleep, climbing distance, survival following oxidative damage. findings behavioral were more efficacious promoting health maintenance life extension; moreover, indicated these effects dose-dependent. Stigmasterols, have restored age-associated decreases steroid hormone levels. Notably, molecular docking predicted promote stigmasterol's binding receptor ECR due its similarity hormones. addition, stigmasterols triggered signaling pathway increasing activity key genes Eip75B Br 20E Jhamt, HmGR, Met, Kr-h1 JH signaling.
