Significance Volume regulation is key in maintaining important tissue functions, such as growth or healing. In this process, the role of efflux cellular fluids difficult to capture due lack apt technologies. Here, we use a tool based on Brillouin light scattering (BLS) that uses interaction laser with inherent picosecond timescale density fluctuations sample. We induced gradual volume decrease multicellular spheroids using osmotic perturbations. BLS revealed nonlinear increase compressibility subsequent increased biopolymer crowding within cells. Our findings should inspire research regarding and tissues stimulate emergence models for cell mechanics at short timescales.

Abstract While the involvement of actin polymerization in cell migration is well-established, much less known about role transmembrane water flow motility. Here, we investigate influx a prototypical migrating cell, neutrophil, which undergoes rapid, directed movement to sites injury and infection. Chemoattractant exposure both increases volume potentiates migration, but causal link between these processes not known. We combine single measurements genome-wide CRISPR screen identify regulators chemoattractant-induced neutrophil swelling, including NHE1, AE2, PI3K-gamma, CA2. Through NHE1 inhibition primary human neutrophils, show that swelling necessary sufficient for potentiation following chemoattractant stimulation. Our data demonstrate chemoattractant-driven complements cytoskeletal rearrangements enhance speed.

Inflammasomes are multiprotein platforms that control caspase-1 activation, which process the inactive precursor forms of inflammatory cytokines IL-1β and IL-18, leading to an type programmed cell death called pyroptosis. Studying inflammasome-driven processes, such as pyroptosis-induced swelling, under controlled conditions remains challenging because signals activate pyroptosis also stimulate other signaling pathways. We designed optogenetic approach using a photo-oligomerizable inflammasome core adapter protein, apoptosis-associated speck–like containing caspase recruitment domain (ASC), temporally quantitatively manipulate activation. demonstrated inducing light-sensitive oligomerization ASC was sufficient recapitulate classical features inflammasomes within minutes. This system showed there were two phases swelling during offers avenues for biophysical investigations into intricate nature cellular volume plasma membrane rupture death.

The presence of impermeant molecules within a cell can lead to an increase in volume through the influx water driven by osmosis. This phenomenon is known as Donnan (or Gibbs-Donnan) effect. Animal cells actively transport ions counteract effect and regulate their volume, pumping Na+ out K+ into cytosol using Na+/K+ ATPase (NKA) pump. pump-leak equations (PLEs) are system algebraic-differential model membrane potential, ion (Na+, K+, Cl-), flux across membrane, which provide insight how combination passive fluxes active contribute stabilizing volume. Our broad objective analytical PLEs three lines investigation: (1) we show that provision extracellular stabilize cell; (2) demonstrate mathematical form NKA pump not important stoichiometry for stabilization; (3) investigate interaction between cation-chloride co-transporters (CCCs) on stabilization, showing NCC destabilize while NKCC KCC it.
