However, little known about psychological well-being survivors US. Objective To explore lived experiences fear cancer recurrence among melanoma. Design, Setting, Participants This was a qualitative survey-based study used semistructured interviews Fear Cancer Recurrence Inventory short form (FCRI-SF) survey tool participants recruited from an academic dermatology practice affiliated University Texas, Austin. Interviews were completed via telephone or person August 2021 to September 2022. Each 9 items FCRI-SF rated on 5-point Likert scale, scored 0 4, maximum possible score 36 points. Data analyses performed February 2022 June 2023. Main Outcomes Measures Semistructured analyzed for themes subthemes The scores tabulated, 13 greater identifying potential cases clinically significant recurrence. Results In all, 51 (mean [SD] age, 49.5 [11.7] years; 34 [67%] female 17 [33%] male) history (stage 0-IIA) interview survey. Among them, (33%) had survived diagnosis stage melanoma, remainder, at least 1 invasive I-IIA). revealed several themes: (1) emotions surrounding follow-up appointments, (2) intensity surveillance, (3) lifestyle changes regarding sun exposure, (4) thoughts life death. Thirty-eight above threshold clinical Conclusions Relevance found despite having excellent prognosis, some even those who 0, have rates intense survivorship affect their well-being.

Introduction Melanoma, a highly aggressive malignancy characterized by rapid metastasis and elevated mortality rates, predominantly originates in cutaneous tissues. While surgical interventions, immunotherapy, targeted therapies have advanced, the prognosis for advanced-stage melanoma remains dismal. Globally, incidence continues to rise, with United States alone reporting over 100,000 new cases 7,000 deaths annually. Despite exponential growth of tumor data facilitated next-generation sequencing (NGS), current analytical approaches emphasize single-gene analyses, neglecting critical insights into complex gene interaction networks. This study aims address this gap systematically exploring immune regulatory dynamics progression. Methods We developed bidirectional, weighted, signed, directed topological network compare transcriptional landscapes between benign melanocytic nevi melanoma. Advanced analysis tools were employed identify structural disparities functional module shifts. Key driver genes validated through centrality metrics. Additionally, deep learning models implemented predict drug-target interactions, leveraging molecular features derived from analyses. Results Significant divergences emerged networks, dominant modules transitioning cell cycle regulation lesions DNA repair migration pathways malignant tumors. A group genes, including AURKA, CCNE1, APEX2, EXOC8, identified as potential orchestrators microenvironment remodeling during transformation. The framework successfully predicted 23 clinically actionable drug candidates targeting these drivers. Discussion observed shift invasion-related provides mechanistic progression, suggesting early therapeutic machinery might mitigate metastatic potential. hub particularly AURKA DDX19B, represent novel immunomodulatory interventions. Summary The strength of evidence supporting a 2‐step wide local excision (WLE) with 5 mm safety margins for melanoma in situ non‐lentigo maligna (non‐LM) type, compared narrower margins, is unclear. This review aims to compare the frequency recurrence (LR) different after complete surgical (R0) non‐LM MIS. We performed systematic literature search PubMed, Scopus, and Cochrane Library up March 17, 2024. PRISMA checklist was used. Of 3,047 articles retrieved, seven retrospective studies were included, enrolling total 1,526 MIS cases excised clear WLE (6 studies) or Mohs surgery (1 study). Most located on trunk/extremities (68%–100%). Narrower used four studies, ranging from no 4 mm, there only one LR. Standard wider remaining three reporting two median follow‐up ranged 48 months 6.6 years. overall certainty quality very low. These findings our highlight that current guidelines recommending re‐excision initial lack strong support this practice.

The incidence of melanoma in situ (MIS) has increased significantly over the past decades, and traditional guidelines for treatment MIS have been excision with a 5-mm clinical margin; however, current Australian other now recommend 5- to 10-mm margins. This changed recommendation was largely driven by outcomes studies using Mohs micrographic surgery, recent surgery are advocating even wider excisions up 18 mm clearance.

Associations all-cause cause-specific were examined as adjusted hazard ratios. The exposures inversely associated all-cause, cardiovascular disease (CVD) cancer mortality.
