Abstract Among the causative agents of neonatal diarrhoea in calves, two most prevalent are bovine coronavirus (BCoV) and intracellular parasite Cryptosporidium parvum . Although several studies indicate that co-infections associated with greater symptom severity, host–pathogen interplay remains unresolved. Here, our main objective was to investigate modulation transcriptome HCT-8 cells during single BCoV C. For this, were inoculated (1) alone, (2) alone , (3) simultaneously. After 24 72 h, harvested analyzed using high-throughput RNA sequencing. Following differential expression analysis, over 6000 differentially expressed genes (DEGs) identified virus-infected co-exposed at hpi, whereas only 52 DEGs found -infected same time point. Pathway (KEGG) gene ontology (GO) analysis showed mostly immune pathways (such as NF-κB, TNF-α or, IL-17), apoptosis regulation transcription, a more limited effect exerted by observed co-infection apparently dominated virus, 800 uniquely hpi. Natural products and herbal medicine have been widely used in drug discovery for treating infectious diseases. Recent outbreak of COVID-19 requires various therapeutic strategies. Here, we YSK-A, a mixture three components Boswellia serrata, Commiphora myrrha, propolis, to evaluate potential antiviral activity against SARS-CoV-2. We showed that YSK-A inhibited SARS-CoV-2 propagation with an IC50 values 12.5 µg/ml 15.42 Vero E6 Calu-3 cells, respectively. Using transcriptome analysis, further demonstrated modulated host gene expressions cells. Among these, selected 9 antiviral- or immune-related genes study. By siRNA-mediated knockdown experiment, verified MUC5AC, LIF, CEACAM1, GDF15 were involved YSK-A. Therefore, silencing these nullified YSK-A-mediated inhibition propagation. These data indicate displays anti-SARS-CoV-2 by targeting multiple genes.
