Extracellular vesicles (EVs) are promising tools for nanomedicine and nanobiotechnology. The purification of mammalian-derived EVs involves intensive processes, their therapeutic application raises multiple safety regulatory issues. Plants have the potential to serve as nonconventional sources therapeutically relevant EVs. In this context, we recently identified hairy roots (HRs) medicinal plants a novel biotechnological platform produce human health.

Abstract Background One of the most effective strategies to increase phytochemicals production in plant cultures is elicitation. In present study, we studied effect abiotic and biotic elicitors on growth, key biosynthetic genes expression, antioxidant capacity, phenolic compounds content Rhizobium (Agrobacterium) rhizogenes -induced hairy roots Ficus carica cv. Siah. Methods The included methyl jasmonate (MeJA) as elicitor, culture filtrate cell extract fungus Piriformospora indica were prepared use. F. exposed elicitores at different time points. After elicitation treatments, collected, evaluated for growth index, total (TPC) flavonoids (TFC) content, activity (2,2-diphenyl-1-picrylhydrazyl, DPPH ferric ion reducing power, FRAP assays), expression level phenolic/flavonoid biosynthesis genes, high-performance liquid chromatography (HPLC) analysis some main comparison control. Results Elicitation positively or negatively affected activities depending elicitor concentration exposure time. maximum chalcone synthase ( CHS : 55.1), flavonoid 3′-hydroxylase F3’H 34.33) transcription factors MYB3 (32.22), Basic helix-loop-helix (bHLH: 45.73) was induced by MeJA elicitation, whereas phenylalanine ammonia-lyase PAL 26.72) UDP-glucose 3-O-glucosyltransferase UFGT 27.57) obtained after P. also caused greatest gallic acid (5848 µg/g), caffeic (508.2 rutin (43.5 quercetin (341 apigenin (1167 µg/g) compounds. Conclusions This study support that Siah can be considered an biotechnological method improved production, course this approach requires further research.

The plant callus culture technique is an emerging source of bioactive compounds with potential applications in cosmetics and pharmaceuticals. Callus-derived extracts contain high concentrations secondary metabolites significant antioxidant anti-inflammatory properties when elicited. Cannabis sativa L. has been used for its medicinal effects; however, the C. extract (CCE) cosmetic remains unexplored. Callus from was induced vitro using a Murashige Skoog (MS) medium supplemented Thidiazuron (TDZ) naphthalene acetic acid (NAA). analyzed composition high-performance liquid chromatography (HPLC). activity assessed DPPH radical scavenging assay. effects were evaluated lipopolysaccharides (LPS)-stimulated RAW264.7 macrophages by measuring nitric oxide (NO) production, DAF-2 fluorescence intensity, released cytokine levels, protein expression inflammatory mediators via ELISA, Western blot, immunofluorescence assays. CCE demonstrated activity. effectively suppressed LPS-induced NO production reduced pro-inflammatory levels. blot analysis revealed that inhibited NF-κB nuclear translocation while upregulating NRF2-mediated responses. Furthermore, HPLC confirmed presence cannabinoids, which could potentially be associated modulation pathways through endocannabinoid system. This study provides evidence possesses notable properties, making it promising ingredient formulations targeting oxidative stress skin conditions.

Among organisms on Earth, plants have the unique ability to produce a wide variety of biomolecules using soil nutrients, air, and solar energy. Therefore, are regarded as most productive cost-efficient bioreactors among living organisms. Flavonoids, major group secondary metabolites exclusively produced in plants, play crucial roles plant physiology various effects human health. Flavonoids used diverse industries such pharmaceutical, nutraceutical, cosmetics industries. These compounds typically extracted from specific that naturally large amounts target flavonoid for commercial production. However, with increasing demand flavonoids, efforts been made enhance production synthetic biology sustainable microbes or plants. Synthetic has utilized metabolic engineering reconstitute biosynthetic pathways flavonoids at whole-pathway level, thereby enhancing For efficient biology, first all, optimized molecular parts enzymes must be identified selected. The best modules precursors final can then constructed these parts. In this review, we summarize enzyme kinetics natural engineered derived different species provide insight into selection parts, design devices, reconstitution based performance biology.

Recent advancements in plant biotechnology have highlighted the potential of hairy roots as a biotechnological platform, primarily due to their rapid growth and ability produce specialized metabolites. This study aimed delve deeper into root development C. asiatica explore optimization genetic transformation for enhanced bioactive compound production. Previously established lines were categorized based on centelloside production capacity HIGH, MID, or LOW groups. These then subjected meticulous label-free proteomic analysis identify quantify proteins. Subsequent multivariate protein network analyses conducted discern proteome differences commonalities. Additionally, quantification rol gene copy numbers was undertaken using qPCR, followed by expression measurements. From analysis, 213 proteins identified. Distinct differences, especially between line other lines, observed. Key related essential processes like photosynthesis metabolism Notably, biomarkers, such Tr-type G domain-containing alcohol dehydrogenase, found HIGH group. The presence ornithine cyclodeaminase emerged significant biomarker linked with indicating successful Rhizobium -mediated transformation. However, qPCR results showed an inconsistency levels, displaying notably higher expression, particularly rolD gene. unveiled importance traceable capacity. strong correlation this emphasizes its role optimizing .
