Multiple sclerosis (MS) and inflammatory bowel disease (IBD) are both autoimmune disorders caused by dysregulated immune responses. Still, there is a growing awareness of the comorbidity between MS IBD. However, shared pathophysiological mechanisms these two diseases still lacking. RNA sequencing datasets (GSE126124, GSE9686, GSE36807, GSE21942) were analyzed to identify differential expressed genes (DEGs) for IBD experimental allergic encephalomyelitis (EAE). Other (GSE17048, GSE75214, GSE16879) downloaded further verification analysis. Shared pathways regulatory networks explored based on DEGs. The single-cell transcriptome central nervous system (CNS) cells sequenced from EAE brains public (PRJCA003980) characteristics Mass cytometry time-of-flight (CyTOF) peripheral blood mononuclear (PBMCs) was performed systematic response in model. Machine learning algorithms also used diagnostic biomarkers MS. We identified 74 common DEGs selected datasets, data intestinal tissues patients showed that 56 highly enriched IL1B+ macrophages. These DEGs, defined as inflammation-related (IRGs), pro-inflammatory macrophages mice patients. abundance CD14+ monocytes validated CyTOF data. IRGs response, NOD-like receptor signaling pathway, IL-18 other related pathways. In addition, 'AddModuleScore_UCell' analysis (such IL1B, S100A8, factors) mainly macrophages, which play an essential role activation multiple sclerosis, such IL-17 NF-kappa B TNF pathway. Finally, suppressors cytokine 3(SOCS3) formyl peptide 2(FPR2) potential machine learning. Two compared control, experiments revealed SOCS3 FPR2 samples. IRGs, encode cytokines, exhibit high expression levels may significant storm biomarkers, FPR2, screened out with great value

Recently, four meta-analyses have explored the association between inflammatory bowel disease (IBD) and risk of stroke. These studies demonstrated that people with IBD may be at an increased However, some limitations such as high heterogeneity lack uniformity in types research, especially reuse sample sizes, cannot neglected. factors reduce credibility their research conclusions. Therefore, we conducted a meta-analysis to explore this possible association.PubMed, Embase, Web Science were searched from inception 30 June 2023. A random effects model generic inverse variance method was used meta-analysis. The Review Manager software obtain all relative risks (RRs) 95% confidence intervals (CIs). Publication bias tested, sensitivity subgroup analyses heterogeneities. This included 12 cohort (involving 4,495,055 individuals). Meta-analysis these data has shown associated stroke (RR = 1.19, 95%CI:1.14-1.24, p < 0.00001). Our results stable robust analyses. Our suggest is To incidence stroke, patients are encouraged undergo assessments, for young female patients; assessing ischemic particular importance. Prospective considering subtypes, severity treatments, regions, other confounding needed further nature each association.https://www.crd.york.ac.uk/PROSPERO/, identifier CRD42022373656.

Objective Previous studies reported possible connections between inflammatory bowel disease (IBD) and several neurodegenerative disorders. However, the comprehensive relationships IBD various disorders were not summarized. We executed a meta-analysis of longitudinal to provide an estimate strength two-directional prospective association Methods accomplished thorough bibliographic search PubMed, Web Science, Embase, PsycINFO, Cochrane Library databases until June 2023 locate relevant studies. The extracted data then analyzed via using either fixed or random effects model. Results final analysis encompassed 27 Individuals with faced increased risk developing four than general public, namely, Alzheimer’s (hazard ratio[HR] = 1.35, 95% confidence interval [CI]: 1.03–1.77, P=0.031), dementia (HR =1.24, CI: 1.13–1.36, P<0.001), multiple sclerosis =2.07, CI:1.42–3.02, P<0.001) Parkinson’s =1.23, CI:1.10–1.38, P<0.001). Two articles incidence amyotrophic lateral system atrophy in patients. Three investigated IBD, revealing elevated latter patients former. (HR=1.87, CI:1.66–2.10, Interpretation These findings verified relationship brain-gut axis, specifically demonstrating heightened diseases among It may be profitable prepare screening strategies for find during long-term course treatment view potential earlier diagnosis diseases, reducing public health social burden.
