Kaposi's sarcoma-associated herpesvirus (KSHV) is a variety of the human gamma-herpesvirus that often leads to occurrence malignant tumors. In addition, sarcoma major cause death among AIDS patients. Ganciclovir (GCV) most widely used drug against KSHV infection in clinic. GCV can restrict vivo synthesis DNA polymerase KSHV, thereby inhibiting replication herpesvirus. However, still suffers from poor specificity and transmembrane capabilities, leading many toxic side effects. Therefore, developing delivery system increases concentrations target cells remains significant clinical challenge. this study, zeolite imidazole salt framework-8 (ZIF-8), biocompatible porous material constructed by coordinating zinc ions 2-methylimidazole, was load GCV. A nano-delivery with microneedle structure also using polydimethylsiloxane (PDMS) mold fabricate MN/GCV@ZIF-8 arrays. These arrays not only offered good skin-piercing capabilities but significantly inhibited cleavage virus vivo, exerting an anti-KSHV function. For these reasons, were able penetrate skin's stratum corneum at tumor site deliver play role.
