Abstract Atherosclerotic cardiovascular disease (ASCVD) has become the leading cause of death worldwide, and early diagnosis treatment atherosclerosis (AS) are crucial for reducing occurrence acute events. However, AS is challenging, oral anti‐AS drugs suffer from limitations like imprecise targeting low bioavailability. To overcome aforementioned shortcomings, Cur/MOF@DS developed, a nanoplatform integrating by loading curcumin (Cur) into metal−organic frameworks with nanozymes magnetic resonance imaging (MRI) properties. In addition, surface‐modification dextran sulfate (DS) enables PCN‐222(Mn) effectively target scavenger receptor class A in macrophages or foam cells within plaque region. This employs mechanisms that scavenge excessive reactive oxygen species microenvironment, promote macrophage autophagy regulate polarization to realize lipid regulation. vivo vitro experiments confirm this outstanding MRI performance effects, which may provide new option AS.

Consequently, exploration multi-targeted safe medications for treating preventing ASCVD has become an increasingly popular trend. Gallic acid (GA), a natural secondary metabolite found in various fruits, plants, nuts, demonstrated potentials ASCVD, addition to its known antioxidant anti-inflammatory effects. Curcumin (CUR) is highly promising for topical therapeutic applications, but water-insolubility one of the major challenges plaguing its drugability, while conventional lipid nanocarriers are limited by low drug-carrying capacity, many additives, and complex processes. In current work, we constructed a composite carrier integrated with cyclodextrin metal-organic framework (γ-CD-MOF) nanosponge (β-CDNS), in which γ-CD-MOF had 13.9 % drug loading 267.1-fold increase solubility water CUR, β-CDNS showed bioadhesion further increasing solubility. The (γ-CD-MOF@β-CDNS) significantly improved vitro release transdermal permeation absorption properties excellent create an advantage local administration treating diseases high exudate, prevents affected area from deteriorating condition counter-absorption formulation.
