Fluorescence navigation is a novel technique for accurately identifying hepatocellular carcinoma (HCC) lesions during hepatectomy, enabling real-time visualization. Indocyanine green-based fluorescence guidance has been commonly used to demarcate HCC lesion boundaries, but it cannot distinguish between benign and malignant liver tumors. This review focused on the clinical applications limitations of indocyanine green, as well recent advances in fluorescent probes fluorescence-guided surgery HCC. It covers traditional imaging such enzymes, reactive oxygen species, sulfur pH-sensitive probes, followed by an introduction aggregation-induced emission probes. Aggregation-induced exhibit strong fluorescence, low background signals, excellent biocompatibility, high photostability aggregate state, show no dilute solutions. Design strategies these may offer insights developing identification surgery.

The challenge of "false positive" signals significantly complicates tumor localization and surgical resection, which are pivotal for successful surgeries. Therefore, the development a method preoperative intraoperative margin determination holds considerable promise improving outcomes. In this study, zero-crosstalk ratiometric tumor-targeting near-infrared (NIR) fluorescent probe was developed precise cancer diagnosis navigation via NIR fluorescence imaging. This integrates moiety that selectively homes in on hepatocellular carcinoma cells exhibits highly sensitive response to γ-glutamyltranspeptidase (GGT), characterized by substantial emission shift from 830 650 nm. unique property enhances its effectiveness early imaging-guided surgery resection. Furthermore, successfully monitors GGT activity blood, cells, vivo, endowing potential clinic. Due efficient targeting situ both subcutaneous tumors orthotopic hepatic tumors, accurate detection capability tumors. Additionally, leveraging imaging, can serve as sprayable tool delineating margins with precision, thereby furnishing real-time imaging guidance during resection surgery.

Abstract Photodynamic therapy is a highly recommended alternative treatment for solid tumors, such as cutaneous or luminal in clinical practice. However, conventional photosensitizers (PSs) often induce undesirable phototoxic effects because of their normal tissue distribution and reduction antitumor resulting from aggregation‐caused quenching effects. The present study developed novel nano‐formulated aggregation‐induced emission (AIE)‐characteristic PS, nab‐TTVPHE, which composed human serum albumin carrier TTVPHE therapeutic agent, more effective cancer with lower Notably, the reactive oxygen species generated by were shielded nanoaggregate structure, photodynamic activity was after nanostructure dissociation. Therefore, this AIE‐characteristic PS provides an innovative strategy effect ability boost potential immunity GSDME‐mediated pyroptosis.
