Abstract Background According to traditional Chinese medicine (TCM), drugs supplementing the vital energy, Qi, can eliminate tumors by restoring host immunity. The objective of this study is investigate underlying immune mechanisms anti‐tumor activity associated with Qi‐supplementing herbs, specifically paired use Huangqi and Danggui. Methods Analysis compatibility regularity was conducted screen combination TCMs. Using MTT assay a transplanted tumor mice model, effects TCMs were investigated in vitro vivo. High content analysis flow cytometry then used evaluate cellular immunity, followed network pharmacology molecular docking dissect significant active compounds potential mechanisms. Finally, mechanism ingredients verified experiments. Results There an optimal Danggui that, administered as aqueous extract, activate immunity suppress more effective than each drug on its own Based analysis, PIK3R1 core target for combined Molecular shows 6 components extract (quercetin, jaranol, isorhamnetin, kaempferol, calycosin, suchilactone) that bind PIK3R1. Jaranol most important component against breast cancer. suchilactone/jaranol and, especially, suchilactone/kaempferol are key enhancement extract. Conclusions cancer individual alone.

Background: Breast cancer is the leading cause of cancer-related deaths among women worldwide. Deciphering molecular mechanisms breast crucial for developing targeted therapeutic approaches. Methods: This study analyzed gene expression profiles from Gene Expression Omnibus (GEO) database to identify differentially expressed genes (DEGs) in cancer. Mendelian randomization (MR) analysis was then employed using publicly available eQTL databases evaluate potential causal relationships between these DEGs and Enrichment analyses were further conducted explore their functional significance. Furthermore, external validation co-expressed The Cancer Genome Atlas (TCGA) database. In vitro assays drug sensitivity performed on selected target validate roles pathogenesis treatment. Results: A total 1052 upregulated 1380 downregulated identified Additionally, MR revealed 12 significant potentially contributing pathogenesis. These primarily enriched lipid metabolism immune responses via regulating microRNA functions AMPK signaling. Validation through TCGA confirmed differential tissues. Strikingly, less-reported DNASE2 ATOH8 demonstrated involvement modulating proliferation, migration, invasion cells. Notably, several commonly used clinical drugs management, such as 5-Fluorouracil, exhibited dramatically increased expression. Conclusions: Our provides novel insights into basis identifies promising strategies this condition.
