Introduction Developing sustainable hydrogen production is critical for advancing renewable energy and reducing reliance on fossil fuels. Cyanobacteria, which harness solar through photosynthesis, provide a promising biological platform generation. However, improving yields requires strategic metabolic genetic modifications to optimize flow overcome photosynthetic limitations. Methods Four cyanobacterial species were evaluated their capacities under varying experimental conditions. Photosynthesis was partially inhibited using distinct chemical inhibitors, including 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU). Exogenous glycerol introduced as supplementary carbon source. Hydrogen monitored over time, rates normalized chlorophyll content. Genomic analysis of transporter proteins conducted identify potential loci further enhancement output. Results Nitrogen-fixing Dolichospermum sp. exhibited significantly higher compared the other tested species. Supplementation with notably increased both rate duration evolution, far exceeding previously established benchmarks. The maximum reached 132.3 μmol H₂/mg Chl a/h—representing 30-fold observed DCMU. screening revealed key putative roles in uptake metabolism. Discussion These findings underscore cyanobacteria, particularly sp., robust platforms production. substantial improvements yield highlight importance targeted engineering supplementation strategies. Future work focused optimizing identified refining interventions could enhance biohydrogen efficiency. By leveraging inherent machinery this offers source significant promise global sustainability.
