Benthic prokaryotic communities, utmost important for wetlands and marine environments functioning, are influenced by physical-chemical parameters interactions with other especially micro-eukaryotes meiofauna. Thus, a holistic view of the benthic community is necessary to fully understand their organization functioning. This study assesses implementation comprehensive view, using mock communities environmental samples. A DNA extraction strategy combining two procedures proposed: one obtain from micro-organisms, 0.25 g sediment, meiofauna, sieving refluxes 5 sediment. Three conditions were considered create communities: (i) varying eukaryotes' abundance, (ii) adding meiofauna salted or freshwater wetlands, (iii) including not sediment matrix. Most organisms composing detected, except filamentous cyanobacteria. All showed similar composition indicating that did affect extraction. result also demonstrated sieving, enrich does significantly any communities. For samples investigated, most taxa usually described in literature retrieved salted, brackish marshes The proposed approach was successful analysing three domains life unique sample, providing community. Furthermore, significant differences observed between different marshes, indicated our can be used conducting ecological studies, useful understanding communities' interactions.
