Rapid and cost-effective tests for the evaluation of industrial municipal effluents are urgently needed environmental monitoring. In this context, peroxidase (PER) activity has been proposed as an early-warning biosensor assessing water quality various wastewater discharges leachates. The peroxidase-toxicity (Perotox) assay includes 0.1 µg/mL PER, albumin, DNA (for protection index), 0.001% monounsaturated Tween-80, substrates luminol H2O2. results revealed that initial burst luminescence was followed by a steady decrease in within first minute, accompanied periodic (cyclic) changes intermediate compound III (CIII) PER. When urban were added, PER inhibited, with concomitant increase lipid peroxidation, indicating oxidative damage. reduction also associated collapse formation CIII, alongside CIII over time. addition to reaction mixture helped mitigate inhibition certain effluents, enabling calculation index. levels polystyrene (PS) organic fraction higher primary aeration lagoon (36 µg/L) compared secondary lagoons membrane filtration (< 16 µg/L). Data analysis negatively correlated population size (r = -0.34) PS materials -0.56). conclusion, Perotox is rapid screening tool identifying potentially toxic complex mixtures, such effluents.
