Abstract The circadian clock system orchestrates daily behavioral and physiological rhythms, facilitating adaptation to environmental internal oscillations. Disruptions in rhythms have been linked increased susceptibility various diseases can exacerbate existing conditions. This review delves into the intricate regulation of diurnal gene expression cell function by clocks across diverse tissues. . Specifically, we explore rhythmicity expressions, behaviors, functions both immune non‐immune cells, elucidating regulatory effects mechanisms imposed clocks. A detailed discussion is centered on complex regulating key cellular signaling pathways. We further diseases, with a focus inflammatory cancers, systemic diseases. By highlighting intimate interplay between especially through clock‐controlled function, this contributes development novel disease intervention strategies. enhanced understanding holds significant promise for design targeted therapies that exploit improved treatment efficacy.

Breast cancer (BC) is one of the most common and fatal malignancies among women worldwide. Circadian rhythms have emerged in recent studies as being involved pathogenesis breast cancer. In this paper, we reviewed molecular mechanisms by which dysregulation circadian genes impacts development BC, focusing on critical clock genes, brain muscle ARNT-like protein 1 (BMAL1) locomotor output cycles kaput (CLOCK). We discussed how rhythm disruption (CRD) changes tumor microenvironment (TME), immune responses, inflammation, angiogenesis. The CRD compromises surveillance features activities effectors, including CD8+ T cells tumor-associated macrophages, that are important an effective anti-tumor response. Meanwhile, review, discuss bidirectional interactions: age rhythms, aging further increases risk through reduced vasoactive intestinal polypeptide (VIP), affecting suprachiasmatic nucleus (SCN) synchronization, ability to repair damaged DNA, weakened immunity. These complex interplays open new avenues toward targeted therapies combination drugs with chronotherapy potentiate response while reducing progression for better outcomes. Melatonin is involved in various functions such as the timing of circadian rhythms, energy metabolism, and body mass gain experimental animals. However, its effects on adipose tissue lipid metabolism are still unclear. This study analyzes melatonin relative gene expression lipolytic proteins rat mesenteric free fatty acid (FFA) glycerol plasma levels male Wistar rats fed a high-fat (HFD) or maintenance diet. Four groups were established: control, obese, control obese plus 2.3 mg/kg/day tap water. After 11 weeks, animals sacrificed at different times throughout 24 h cycle, samples collected analyzed. Cgi58, Perilipin, Dgat1 expression, well FFA concentrations, showed rhythm patterns group. HFD disrupted those increased concentrations during dark photoperiod. In both melatonin-treated groups, almost all analyzed genes patterns. Notably, significantly prevented increase photoperiod (obese group: ~1100 mM vs. + ~600 μM, similar to levels). rhythmic pattern observed was not sustained. According our results, could regulate transcription lipolysis proteins. The effect preventing elevated associated with diet intake warrants further investigation.

The complex interaction between circadian rhythms and physiological functions is essential for maintaining human health. At the heart of this lies PERIOD proteins (PERs), pivotal to clock, influencing timing behavioral processes impacting oxidative stress, immune functionality, tumorigenesis. PER1 orchestrates cooperation enzyme GPX1, modulating mitochondrial dynamics in sync with daily thus regulating mechanisms managing energy substrates. PERs innate cells modulate temporal patterns NF-κB TNF-α activities, as well response LPS-induced toxic shock, initiating inflammatory responses that escalate into chronic conditions. Crucially, cancer cell behaviors including proliferation, apoptosis, migration by levels cycle stimulating expression oncogenes c-Myc MDM2. PER2/3, antagonists stem biology, play important roles differentiating their stemness. Importantly, Pers serve a significant factor early diagnosis prognosis. This review delves link rhythm regulator PERs, disruptions rhythm, oncogenesis. We examine evidence highlights how dysfunctions activities initiate development, aid tumor growth, modify metabolism through pathways involved stress system. The identified 17 compared day, covering vaccinations against COVID-19 (9), influenza (5), hepatitis B (2), (1), pneumococcal infection (1). Eleven out these demonstrated statistically significant effects response, with 10 reporting stronger morning afternoon vaccination. Of six subgroups an average age 60 years older, five showed significantly vaccination, while sixth only men. In contrast, 16 younger than titers. Chronopharmacokinetics is a specialized field within pharmacokinetics that focuses on how the timing of drug administration affects absorption, distribution, metabolism, and elimination drugs. The circadian rhythm, governed by body's biological clock, plays crucial role in pharmacokinetics, which encompasses These pharmacokinetic processes are subject to daily variations influenced rhythm. Drug absorption can fluctuate due changes gastrointestinal function, such as motility gastric pH, blood circulation. Similarly, distribution drugs body factors tissue perfusion, plasma protein binding, cell membrane permeability, all modulated Metabolism, especially hepatic also experience affect rate at processed converted into active or inactive metabolites. Finally, drugs, primarily through renal show rhythms impact excretion compounds from body. Understanding these fluctuations essential optimize efficacy safety pharmacological treatments, dosages times could be adjusted align with peaks clock activity. Thus, treatments more effective have fewer side effects leveraging variability processes.
