Abstract Malaria poses a serious threat to human life and is prevalent in tropical subtropical areas across the globe. Drugs such as quinine, chloroquine (a synthetic version of quinine), artemisinin, its derivative compounds have been used treat malaria. Developing highly effective chemical scaffolds with minimal toxicity necessary because malarial parasites become resistant existing drugs. In this context, 1,2,4,5-tetraoxanes emerged crucial framework notable antimalarial properties. Objective: The present research aimed to prepare and characterize glyceryl dilaurate (GDL) containing solid lipid nanoparticles (SLN) with tween 80 lecithin as an emulsifier in which the artesunate drug was loaded. Methods: SLNs were synthesized by solvent emulsification–diffusion technique. formulation characterized for size, size distribution, zeta potential, shape morphology, DSC. In vitro release studies performed at pH 5.0 7.4 mimic vivo conditions. Hemolytic vitro, antiplasmodial activities carried out. Plasmodium berghei (NK65 resistant strain) infected mice used test antimalarial efficacy of SLN. Results: SLN exhibited 327 nm average sizes 0.164 PDI and-23.4 mV potential. TEM images revealed a spherical structure. entrapment efficiency ART calculated 85.68%. showed that entrapped released weakly acidic environment (83.45% ART). ART-GDL conjugate stable safe parenteral delivery. IC50 value GDL-based ART-SLNs be 0.32 µM. Furthermore, resulted enhanced parasite killing P. berghei-infected improved survivability compared free administration. Conclusion: allows effective intravenous administration artesunate. Thus GDL-Based could potential delivery system therapy.

An efficient method for the direct synthesis of ozonides via reaction alkenes with an ozone stream under solvent-free conditions is described. In this study, alkene substrates were loaded over a roughly ground molecular sieve and exposed to give corresponding ozonides. The new synthetic protocol was successfully applied various substituted in good yields.
