Abstract Growing environmental concerns and the need to adopt a circular economy have highlighted importance of waste valorization for resource recovery. Microbial consortia-enabled biotechnologies made significant developments in biomanufacturing valuable resources from biomass that serve as suitable alternatives petrochemical-derived products. These microbial consortia-based processes are designed following top-down or bottom-up engineering approach. The approach is classical method uses variables selectively steer an existing consortium achieve target function. While high-throughput sequencing has enabled community characterization, major challenge disentangle complex interactions manipulate structure function accordingly. prior knowledge metabolic pathway possible among partners design engineer synthetic consortia. This strategy offers some control over composition targeted bioprocesses, but challenges remain optimal assembly methods long-term stability. In this review, we present recent advancements, challenges, opportunities further improvement using approaches microbiome engineering. Abstract In microbial systems, a metabolic pathway can be either completed by one autonomous population or alternatively distributed among consortium performing division of labor (MDOL), where several specialized populations cooperate to complete the pathway. MDOL facilitates system’s function reducing burden; however, it may also hinder exchange efficiency intermediates individuals. As result, community is influenced trade-offs between specialization and versatility individuals, with latter potentially introducing redundancy into community. However, remains unclear how individuals involved controlled in order optimize this study, we deconstructed naphthalene degradation four steps introduced them individually combinatorically different strains, varying levels specialization. Using these engineered 1,456 synthetic consortia tested their efficiency. We found that 74 possessing exhibited higher than both rigorous Quantitative modeling derived from our experiments provides general strategies for identifying most effective functional (MCFR) range possible MCFRs. Our large-scale genomic analysis suggests natural communities hydrocarbon are mostly functionally redundant. summary, study critical insights engineering high-performance systems explains why prevalent communities.
